BackgroundAlzheimer’ s disease is one of the most common progressive neuro-degenerative diseases,and the etiology of Alzheimer disease is not yet fully illustrated and the pathogenesis is complex.At present,there still no available western medicine that can stop or reverse the progression of this disease.Traditional Chinese medicine treatment of AD attracts more and more attention.Bushen Yizhi Formula(BSYZF),a Chinese prescription(consist of Fructus cnidii,Chinese wolfberry,Glossy privet fruit,Ginseng,Prepared fleece flower root and Cortex moutan)that have been proved by our department,to be capable of ameliorating the symptoms of Alzheimer’ s Disease(AD)in patients and animal models.Moreover,the drug-containing serum has certain protective effect on AD cells model.We now show that BSYZ-E can improve behavioral performance in the Scopolamine model in mice of AD.ObjectiveTo study the efficacy of BSYZ-E on ameliorating the AD pathology and learning memory ability in Scopolamine model in mice.Futher more,to dectetive whether BSYZ-E can ameliorate the pathology of AD via adjusting the cholinergic system.MethodsThe 8-month-old male Kunming mice were randomly divided into 5 groups:control group,model group,positive group,low-dose BSYZ-E group,high-dose BSYZ-E group,In addition,each group of mice were given the corresponding solution to gavage daily.After 1 weeks,spatial learning and memory were tested by Morris water maze test.And detect the levels of acetyl cholinesterase activity and choline acetyltransferase in brain tissuesResults(1)Results of Morris water mazeCompare with the control group,the escape latency of model group significantly prolong(P<0.01),the times of crossing the position of the platform significantly decrease(P<0.01)and the duration in target quadrant significantly decrease(P<0.01).Compare with the model group,the escape latency of positive group and high-dose group significantly shorten(P<0.01),the times of crossing the position of the platform significantly increase(P<0.05)and the duration in target quadrant of high-dose group significantly increase(P<0.05).(2)The levels of acetyl cholinesterase activity and choline acetyltransferase in brain tissuesCompare with the control group,the levels of AchE in model group significantly increase(P<0.01).Compare with the model group,the levels of AchE in positive group and high-dose group significantly decrease(P<0.05).Compare with the control group,the levels of ChAT in model group significantly increase(P<0.01).Compare with the model group,the levels of AchE in positive group and high-dose group significantly decrease(P<0.05).ConclusionMorris water maze test results suggest BSYZ-E,to a certain extent,can improve the learning and memory abilities of scopolamine model mice,which may via adjusting the cholinergic system.The results show that BSYZ-E may be effective in the treatment for Alzheimer’s disease,and this deserves further research.
