Leukemia is an important disease that threatens human health.Currently,there is no effective treatment for leukemia.Ginseng has been used as a traditional Chinese medicine for the treatment of cancer.Modern medicine has also demonstrated that ginsenosides are active ingredients in ginseng,and ginsenosides can be used to treat a variety of cancers.Ginsenoside Rh2 is a commonly used drug for the treatment of leukemia,however,it has not been reported that the comparison on the treatment of acute myeloid leukemia and chronic myelogenous leukemia and the induction of cell autophagy.In this paper,we established the human acute myeloid leukemia(U937)and chronic myelogenous leukemia(K562)cell models and set different concentrations of ginsenoside Rh2 treatment group to study the proliferation inhibition,apoptosis,autophagy,and related mechanisms of U937 and K562 induced by ginsenoside Rh2.The inhibitory effect of this method on the proliferation of leukemia cells was confirmed by proliferation inhibition experiments.The apoptosis induced by ginsenoside Rh2 was studied by measuring the apoptosis rate,reactive oxygen species,and mitochondrial membrane potential.The results showed that ginsenoside Rh2 can effectively inhibit the proliferation of human acute myeloid leukemia cells(U937)and chronic myelogenous leukemia cells(K562),causing apoptosis through the mitochondrial pathway,and it is better than U937 cells for K562 cells.Autophagy is an important mechanism by which cells maintain homeostasis,but excessive autophagy can cause cell death.In this paper,we also studied the effect of ginsenoside Rh2 on autophagy of leukemic cells U937 and K562 cells.We used fluorescence staining to observe the production of autophagy.We used real-time fluorescence quantitative PCR to study the expression of autophagy-related genes.Western blot was used to study the expression level of autophagy-related proteins.The study found that ginsenoside Rh2 can induce autophagy production in U937 and K562 cells,and the cells induced in K562 cells have the strongest autophagy.The study of this paper provided an important basis for the treatment of leukemia with ginsenoside Rh2,and laid the foundation for studying the effect of ginsenoside Rh2 on leukemia cell apoptosis and autophagy.
