| Objective: To study the pathological changes of endoplasmic reticulum stress and apoptosis in animal brain tissue after cardiac arrest(CA)and cardiopulmonary resuscitation(CPR).Methods: Healthy SD rats were randomly divided into sham(SH)group,normal saline(NS)group,the tea polyphenols(TP)group,in addition to the SH group,others were induced ventricular fibrillation esophagus stimulation with alternating current.Five min after cardiac arrest CPR was performed.Once restoration of spontaneous circulation(ROSC)was gained,TP or NS were immediately administrated.Cortex of rats in each group were collected at 12,24,48,72 h after ROSC.Tunel(TdT mediated DUTP Nick end labeling)and caspase3、CCAAT/enhancer-binding protein homologous protein(CHOP)fluorescence intensity,as well as caspase12 and molecular chaperone78 KDa glucose-regulated protein(GRP78)expression of western bloting at each time point were measured respectively.Results: 1.Tunel and caspase3 fluorescence density at 72 h of TP groupand NS group were greater than SH group(p< 0.05),while the Tunel and caspase3 fluorescence density of TP group were smaller than that of NS group(p< 0.05);caspase12 expression of TP at 48,72 h decreased significantly when compared with NS group(p < 0.05),and the expression of NS group and TP group were higher than that in SH group(p<0.05).2.Compared with SH group,the expression of GRP78 in NS group and TP group was significantly higher in different time(p< 0.05),The expression of GRP78 in the TP group was significantly higher than that in NS group in different time(p < 0.05).Compared with SH group,the express of CHOP in TP group and NS group were significantly higher at 48,72 h.however,the expression level of CHOP in TP group was lower than NS group(p< 0.05).Conclusions: TP can reduce the activation of caspase-12 and caspase-3induced by ischemia reperfusion injury and inhibit the apoptosis of neural cells,which is related to the down-regulation of ER stress. |
