| BackgroundAdenosine is an endogenous nucleoside that can exert multiple biological effects in the human body.It activates four G protein-coupled receptors(A1R,A2 aR,A2bR,A3R)allows tissues and cells to adapt to these damage under energy consumption and ischemia and hypoxia.Mitochondria are the key subcellular targets for neuronal cell death after cerebral ischemia.The survival of neurons is closely related to the functional integrity of mitochondria.Therefore,the key to protect the neurons after cerebral ischemia is to protect mitochondria.In the past,the effect of adenosine on the mitochondrial function of ischemia-reperfusion injury has not been reported in the literature for the time being.Therefore,it is very meaningful to explore whether adenosine has a protective effect on brain tissue from the perspective of mitochondria.This study was to observe the effect of adenosine on mitochondrial function after cerebral ischemia-reperfusion injury and its possible mechanism.ObjectiveObserve the effect and related mechanism of adenosine preconditioning on mitochondrial function after focal cerebral ischemia-reperfusion injury in rats.MethodForty-two male Sprague Dawley(SD)rats were randomly divided into three groups according to the principle of random sampling: sham operation group(sham group),ischemia reperfusion group(IR group),adenosine preconditioning In the treatment group(adenosine pretreatment group,AP group),14 rats in each group,ischemia reperfusion group middle cerebral artery occlusion(MCAO)models were prepared in IR group and AP group respectively.after 2 hours of successful modeling,pull the thread 1cm,After 22 hours of reperfusion,neurological function scores were given in each group.Infarct size was measured by TTC staining in each group.Rat mitochondrial membrane potential,mitochondrial matrix calcium ion concentration,ATPase activity,and ATP content were also observed in 10 rats of each group.Result1.Both the ischemia-reperfusion group and the adenosine group had neurological deficits.However,The neurological deficit score of the adenosine group was lower than that of the ischemia-reperfusion group,and the difference was statistically significant(P<0.05).2.Compared with sham group,the mitochondrial matrix calcium concentration in IR group was significantly higher,and mitochondrial membrane potential,ATPase activity,and ATP content were significantly decreased,and the difference was statistically significant(P<0.05).3.The adenosine pretreatment significantly reduced the mitochondrial matrix calcium concentration and increased the mitochondrial membrane potential,ATPase activity and ATP content,and the difference was statistically significant(P<0.05).Conclusion1.Adenosine has a protective effect on focal cerebral ischemia-reperfusion injury in rats,which reduces the neurological impairment in rats.2.Adenosine preconditioning significantly improved mitochondrial membrane potential,increased ATP content,increased ATPase activity,and decreased mitochondrial matrix calcium concentration in rat ischemia-reperfusion models,which may protect mitochondrial function through these mechanisms. |
