| Objective:To evaluate the differential expression of mRNA and miRNA in epidermal cells of guinea pigs following infection with HSV2 and investigate the expression of toll receptor pathway mRNA and miRNA in epidermal cells of guinea pigs infected with HSV2 and guinea pigs treated with Longdanxiegan granules.Methods:Establishment of model infected with HSV2 of female guinea pigs.The guinea pigs were divided into randomly three groups(n=15):Longdanxiegan group,ACV group,genital herpes group,and 15 normal female guinea pigs were used as controls.The genital epithelial cells of guinea pigs in the genital herpes and normal control groups were harvested,RNA was isolated and cryopreserved.After being treated with Longdanxiegan Granules and ACV,the vulvar epithelial cells were harvested from the Longdaxiegan group and the ACV group.The RNA was isolated and then the RNA was sequenced and analyzed.Observe the expression pattern of mRNA and miRNA in epidermal cells of female guinea pigs infected with HSV2 and its expression differences with the normal group,as well as the relationship between mRNA and miRNA expression.Observe the changes of TLR pathway-related mRNA and miRNA expression in epidermal cells of guinea pig guinea pigs infected with HSV-2 by Longdanxiegan granules.Results:1.A total of 424 miRNAs and 9,336 genes were sequenced In the present study and 364 miRNAs and 4,130 genes were identified following data filtration and genome annotation.A total of 105 mi RNAs were upregulated and 141 were downregulated;whereas 798 genes were upregulated and 503 were down-regulated inthe HSV2-infected group,as compared with the control group.2.Aterations in the TLR pathway after infection with HSV2.Genes related to the TLR pathway are under the regulation of miRNAs.and the negative correlation between miRNA and mRNA of these genes.the expression levels of TLR1,TLR3,TLR5,TLR9,TRAF6 and TRIF significantly decreased following infection with HSV2(P <0.05);whereas the expression levels of the mi RNAs,including miR-592,miR-1245b-5p,miR-150,miR-342-5p,miR?1245b?3p and miR?124,significantly increased(P <0.05).3.The expression levels of TLR1,TLR3,TLR5,TLR9,TRAF6 and TRIF significantly increased after treatment with Longdanxiegan Granules(P <0.05);whereas the expression levels of the mi RNAs,including miR-592,mi R-1245b-5p,miR-150,miR-342-5p,miR?1245b?3p and miR?124,significantly decreased(P<0.05).And Longdanxiegan granules treatment effect is significantly better than ACV.Conclusion:The expression of mRNA and mi RNA in epidermal cells of guinea pigs infected with HSV2 was different,and the negative correlation between miRNA and mRNA of these genes.Longdanxiegan Granules interfered with mRNA and miRNA related to TLR pathway in epidermal cells of guinea pig infected with HSV-2. |
