Objective:To observe the influence of moxa cone moxibustion on learning and memory ability,expression of Insulin in blood and cerebral tissue in D-galactose induced aging mice,to explore the mechanism and provide experiment evidence for clinic application.Methods:60 male mice(3 months old)were randomly and equally divided into the normal sodium control group,model group,moxa cone moxibustion 1 group(acupoint ST36 and GB39),moxa cone moxibustion 2 group(acupoint DU20 and RN4)and moxa cone moxibustion3 group(nonacupoint).Rats in treatment and model groups were given with D—galactose injection for 70 days,while rats in the normal sodium control group were injected the same dosage of normal sodium 12 days later,rats in the treatment groups were treated for every day for 58 days.Immunohis to dosemistry was used to detect the SOD2 and GADD45,The real-time fluorescence quantitative RT-PCR assays was used to detect the SOD2 and GADD45 mRNA.HE was used to detect Neuronal cell morphology.Results:(1)The Immunohistodosemistry results showed: Compared with the normal sodium control group,SOD2、GADD45 of model group decreased obviously(P<0.05),compared with the model group,moxibustion group 1 and moxibustion group 2 increased,the difference was significant(P<0.05),compared with the moxibustion group 3,moxibustion group 1 and moxibustion group 2 increased,the difference was significant(P<0.05)(2)The RT-PCR results showed:Compared with the normal sodium control group,SOD2mRNA、GADD45mRNA of model group decreased obviously(P<0.05),compared with the model group,moxibustion group 1 and moxibustion group 2 increased,the difference was significant(P<0.05),compared with the moxibustion group 3,moxibustion group 1 and moxibustion group 2 increased,the difference was significant(P<0.05).(3)HE results showed: Compared with the normal sodium control group,necrotic neuron cells of model group increased obviously(P<0.05),compared with the model group,moxibustion group 1 and moxibustion group 2 decreased,the difference was significant(P<0.05),compared with the moxibustion group 3,moxibustion group 1 and moxibustion group 2 decreased,the difference was significant(P<0.05).Conclusions: Moxa cone moxibustion on ST36 and GB39,DU20 and RN4 can both improve the SOD2 and GADD45 of Brain in D-galactose induced aging mice thus preventing brain aging,Yisui moxibustion has antiaging effect.
