| Background and purpose:Epithelial ovarian cancer(EOC)is one of the common malignant tumors in the female reproductive system.By analyzing the cell cycle related protein CENPF expression level in EOC and the relationship between its expression level and EOC clinical features,as well as the function of CENPF on ovarian cancer cell activities,the aim of this study is to evaluate whether CENPF can be used as a biomarker of EOC and to investigate the potential roles of CENPF in the development of EOC.Methods:1.Different databases and immunohistochemistry were used to analyze the expression level of CENPF and the relation between its expression level and EOC prognosis;2.Cell proliferation,colony formation,scratches and transwell assays were performed to investigate the effect of CENPF on the proliferation and migration of ovarian cancer cells.3.The databases were used to predict upstream transcription factors of CENPF,and GSEA analysis were used to identify the possible involved signaling pathways of CENPF.Results:1.Database analysis and immunohistochemistry results showed that CENPF expression in EOC was significantly higher than normal ovarian tissue and its expression was associated with poor prognosis(P value<0.001);2.Proliferation and migration ability of ovarian cancer cell was significantly suppressed by specific knockdown CENPF(P value<0.001);3.NFKB1 and other related transcription factors may regulate CNEPF transcription,which is a cell cycle related gene.CNEPF is mainly involved in the cell cycle pathway by GSEA analysis.Conclusions:1.The expression of CENPF is clearly up-regulated in EOC,and associated with EOC prognosis,pointing out that CENPF has the potential to be a prognostic marker of EOC.2.As a cell cycle related gene,CNEPF has important function on promoting the proliferation and migration of ovarian cancer cells through relevant signaling pathway. |
