| ObjectThe aim of this study was to detect the expression of transient receptor potential vanilloid type 1(TRPV1)in colonic mucosa of patients with irritable bowel syndrome(IBS)by immunohistochemistry,and investigate the relationship between TRPV1 and the clinical symptoms of IBS patients and the possible mechanism.MethodsFrom July 2013 to November 2017,there were 29 cases of IBS patients in our department of the Second Affiliated Hospital of Soochow University included in the experimental group(IBS group)according to the standard of Roman III.16 cases who were examined after physical examination were all negative in colonoscopy as normal control group(N group).The colonoscopy was performed in the IBS group and the N group,and the colonic mucosa was removed with a biopsy forceps to make a paraffin section.The results of routine pathological examination showed normal,and then the expression of TRPV1 was detected by immunohistochemistry.Results1.The expression of TRPV1 in the colonic mucosa of IBS group and N group were 0.8(0-8.4)and 0.4(0-1),with the Mann-Whitney U test,the expression of TRPV1 were significantly higher in colonic mucosa in IBS patients than that in the N group(P<0.05);2.Correlation of the expression of TRPV1 with IBS Symptoms: Spearman correlation was used to determine the relationship between TRPV1 expression and the symptoms of IBS patients.The scatter plot was used to intuitively judge the monotonic relationship between the two.The results showed that there was a positive correlation between the expression of TRPV1 and the symptoms of IBS patients.r= 0.772,P<0.001;3.The expression of TRPV1 in IBS subgroups:1.1(0-8.4)in the IBS-D group,0.6(0.2-6.2)in the IBS-C group,and 0.8(0-7.8)in the IBS-M group;nonparametric tests were used(Kruskal-Wallis test),P=0.938>0.05,so there was no statistical difference in TRPV1 expression levels in IBS subgroups.4.Differential expression of TRPV1 in sexes: Expression in male IBS patients was 1.0(0-7.2),and in female IBS patients was 0.8(0-8.4),using the rank sum test(Mann-Whitney U test),P=0.871> 0.05,indicating that the expression level of TRPV1 in IBS patients was not statistically different between men and woman.5.Differences in IBS patient symptom scores between men and women: The IBS symptom score for male patients was 4.0(2-9.5),and the IBS symptom score for female patients was 3.75(2.0-11.5).The rank sum test(Mann-Whitney U test)was used.P= 0.835>0.05,indicating that the symptom scores of IBS patients are not significantly different between men and women.Conclusion1.The expression of TRPV1 in colonic mucosa of IBS patients was significantly increased,but there was no significant difference among IBS subtypes.2.There is a positive correlation between the expression of TRPV1 and the symptoms of IBS patients;3.There was no statistically difference in the expression of TRPV1 between males and females in IBS patients;4.There was no statistically difference in the IBS symptom scores between men and women;... |
