Objective To establish rat models of diabetic and hyperlipoidemia erectile dysfunction for studying the expression of endothelial nitric oxide synthase(eNOS)and endothelin 1(ET-1),change of penis tissue structure in the corpus cavernosum of rat model,then explore the possible mechanism.Methods 60 male SD rats of SPF grade were randomly divided into three groups:control group’(n=15),high-fat group(n=15)and diabetes group(n=30).control group fed with normal feed,high-fat group and diabetes group fed with lipid rich food for 24 weeks,and then the formor given containing 0.2%oxygen pyrimidine lipid rich food to keep feeding,while the later given 20 mg/kgSTZ by intraperitoneal injection.At the end of 36w,we observed the spontaneous erectile response induced by APO.then Fasting Blood glucose,serum TC,TG,LDL-C,HDL-C and Fasting insulin levels were measured;The expression of eNOS and ET-1 in corpus cavemosum tissue were detected by western blot.Penile tissues were embedding,fixed,HE and masson staining to observe rat corpus cavemosum structure.Results The results of fasting blood glucose,fasting insulin,blood lipid showed and erectile function testing show that the rat model was established successfully;Western blot showed that compared with control rats,the expression of eNOS decreased remarkably,while the ET-1 was incresed significantly(P all<0.05)in high fat and diabetes group,compared with high-fat group,the expression of eNOS and ET-1 further imbalance(Lower expression of eNOS and higher expression of ET-1).HE and masson staining indicated that smooth fibers decreased while collagen fibers increased,The ratio of cavernous smooth muscle/collagen fibers decreased.diabetes mellitus group was the most serious in the three groups.Conclusion the pathogenesis of diabetes and hyperlipidemia indused erectile dysfunction may be closely related to imbalance expression of eNOS and ET-1 in corpus cavemosum tissues and alteration of smooth muscle and collagen fiber structure.
