Lepisorus Ussuriensis(Regel et Maack)Ching,a plant belonging to family Lepisorus of the Polypodiaceae,has the heat-clearing and detoxifying effects.It is widely used in the treatment of urination,hematuria,edema,sore throat,and other diseases.However,there are few reports on its chemical composition and biological activity.In order to further elucidate its chemical composition and screen its biological activity,the following research had been carried out on this medicinal plants grown in Jilin Province.The ICP-MS technology was used to determine the essential trace elements in this plant for the first time.The contents of the elements such as Mn,Zn,Cr,Ni,Cu,V,Mo,Se,Co,Sn were 213.70,48.63,13.89,6.54,6.50,2.57,1.23,0.58,60,0.30 ?g/g respectively.The results showed that the plant was rich in the trace elements essential to human such as Mn,Zn,Cr,Ni,Cu,V.The work should fill the blank about the study of trace elements in L.ussuriensis.2.For the first time,the extraction process of total flavonoids and total phenolic acids from L.ussuriensis was optimized by response surface methodology.The obtained optimal extraction conditions were as follows: ethanol volume fraction was 80%,extraction times was 3,and material-liquid ratio was 1: 30.The contents of total flavonoids and total phenolic acids in the extracts obtained were 15.8% and 8.8%,respectively.The study provided raw materials for further chemical composition and biological activity research.3.A total of 18 compounds were isolated and purified by silica gel column chromatography,preparative thin layer chromatography and semi-preparative HPLC chromatography.There were 15 compounds identified by HR-MS and NMR.They are quercetin 3-O-?-D-galactopyranoside(1),quercetin(2),genistein(3),Ligustilide(4),(3S)-6,8-dihydroxy-3-phenyl-3,4-dihydroisocoumarin(5),?-Spinach sterol(6),Eudesamin(7)caffeic acid(8),Ethylcafeate(9),?-sitosterol(10),Bis-(2-ethylhexyl)-phthalate(11),Di-n-octyl phthalate(12),3,27-dimethyl-Nonacosane(13),7-hydroxycoumarin(14)and chlorogenic acid(15).Among of them,11 compounds including compounds 3,4,5,6,7,8,9,11,12,13,14 were isolated from L.ussuriensis for the first time.4.Classical models such as cough induction by ammonia water and tracheal phenol red output were applied to assess the effect of antitussive and expectorant activities.Furthermore,the acute inflammation model induced by tracheal instillation with LPS was used to investigate the anti-inflammatory mechanism.The results showed that the plant possesses significant antitussive and expectorant activities.And the effect may be occured by decreasing the levels of cytokines such as IL-6,IL-1? and TNF-? in serum.5.UPLC-Q/TOF-MS combined with multivariate statistical analysis were used to study the constituents absorbed in the blood of healthy mice and inflammatory pathological model mice after intragastric administration with 80% ethanol extract of L.ussuriensis.The aim is to clarify the effective components.A total of 22 prototype composition were identified in the healthy mice.While a total of 31 prototype composition were identified in the inflammatory model mice.There are great difference between the healthy and the model mice.Three known components including luteolin-7-O-?-D-glucoside,isoviterine,and phenylethanoid glycoside B were identified in inflammatory model rats.And it was presumed that these 3 compounds were closely related to the anti-inflammatory efficacy.In summary,an in-depth study of the chemical composition,antitussive and expectorant effect and mechanism,components absorbed in blood,were performed.The study could improve the chemical composition research and expand the scope of medicinal use.Most importantly,it could provide the scientific data and theoretical basis for the further development of L.ussuriensis.
