Objective As a new type of gas signal transduction molecule,Hydrogen sulfide is gradually well known.The effect of Hydrogen sulfide is closely related to the opening and closing of ion channels.Among them,KATP is the most closely related.In this study,H9C2 rat cardiomyocytes were cultured under anaerobic conditions to observe the effect of Hydrogen sulfide on apoptosis and the role of KATP in them.At the same time,considering KATP blockers or openers as the treatment factors and exploring the effect of KATP on H2 S,which could find a new intervention target of heart failure clinical treatment with experimental and theoretical basis.Methods The experimental subjects were H9C2 rat cardiomyocytes cultured in hypoxia at 37 ℃,94% N2,1% O2 and 5% CO2.The cells were randomly divided into six groups: 1.blank control group:Conditions for 24 hours.2.Na HS group: 400 μ mol/l Na HS after cardiomyocytes were cultured in hypoxic-ischemic conditions for 24 hours.3.glibenclamide group: 1mmol/l of glibenclamide cardiomyocytes,cultured in hypoxia-ischemia conditions for 24 hours.4.Pinacidil group:50mmol/l pinacidil treatment of myocardial cells,cultured in hypoxia and hypoxia conditions for 24 hours.5.Na HS + glibenclamide group: 400μmol/l of Na HS + 1mmol/l of glibenclamide co-treatment of myocardial cells,cultured in hypoxia and hypoxia for 24 hours.6.Na HS + pinacidil group: 400 μ mol/l Na HS + 1mmol/l Pinacidil Co-treatment of myocardial cells,cultured in hypoxia-ischemia conditions for 24 hours.The above steps are repeated three times.Western blotting was used to detect the expression of Caspase-3 and Caspase-9.The expressions of bcl-2 and bax were detected by immunohistochemistry.Cell viability was detected by MTT assay.Compare the data between groups.Results Compared with the blank control group,Na HS preconditioning reduced the expression of Caspase-3 and Caspase-9,decreased the ratio of Bax / Bcl-2 and increased the cell relative survival rate.Compared with the blank control group,glibenclamide itself had no significant difference on the cell relative survival rate and the expression of Caspase-3 、 Caspase-9 、 Bax and Bcl-2 in cardiomyocytes.Compared with Na HS group,pretreatment with Na HS and glibenclamide increased the expression of Caspase-3 and Caspase-9,the ratio of Bax / Bcl-2 increased and the cell relative survival rate decreased.Compared with Na HS group,pretreatment with Na HS and pinacidil increased the expression of Caspase-3 and Caspase-9,the ratio of Bax / Bcl-2 increased and the cell relative survival rate decreased.Compared with the normal control group,pretreatment with pinacidil alone increased the expression of Caspase-3 and Caspase-9,the ratio of Bax / Bcl-2 increased and the cell relative survival rate decreased.Conclusions Exogenous H2 S inhibits cardiomyocyte apoptosis by activating KATP channel.
