Effect Of Kappa-selenocarrageenan And N-Acetyl-D-Glucosamine On Humoral Immune Response Induced By Hepatitis A Virus In Mice

Objective To investigate the effect of Kappa-Selenocarrageenan and N-Acetyl-D-Glucosamine on humoral immune response induced by hepatitis A virus(HAV)in mice.Methods:Eleven experiment groups are set up,with 7 ICR mouse per group randomly,negative control group(saline),pure antigen group(18EU HAV),Aluminium adjuvant group(AL(OH)3 0.3mg+18EU HAV),Kappa-Selenocarrageenan experimental groups(KSC lmg+18EU HAV,KSC 0.5mg+18EU HAV,KSC 0.25mg+18EU HAV,KSC 0.1mg+18EU HAV),N-Acetyl-D-Glucosamine experimental groups(18EU HAV+GIcNAc0.5mg,18EU HAV+GlcNAc1.0mg,18EU HAV+GlcNAc1.5mg,18EU HAV+GlcNAc2.0mg),in which mice were injected subcutaneously with 300 mL,immunizing one time.The anti-HAV specific IgG antibody levels were tested by indirect ELISA method in the serum of mice after the first 4th,8th,12th,16th week of injection.The healthy statuses of mice were observed during the study.The kidney,spleen,liver,lung and heart of mice immunized with KSC + HAV at the optimal dosage and those in negative control group were taken out at week 16 after immunization,and prepared into sections for pathological observation.The GlcNAc experimental group was compared by observation.Results HAV-specific IgG antibody of all mice were detected at four setting time point except negative control group.The antibody level of all groups increased first and then decreased.The peak of Aluminium adjuvant group in the 12th week and others in the 8th week.The level of antibody in KSC 0.5mg+18EU HAV group at week 8,12 was higher than that in simple antigen group(P<0.05).The level of antibody in KSC 0.25mg+18EU HAV group at week 12 was higher than that in pure antigen group(P<0.05).The level of antibody in GlcNAc1.0mg+18EU HAV group at 4,8,12 weeks was higher than that in simple antigen group(P<0.05;P<0.001).In the 4th week,the level of antibody in GlcNAc 1.0mg+18EU HAV group was higher than that of Aluminium adjuvant group(P<0.05).The optimal dose for this experiment was KSC 0.5 mg,GIcNAc 1.0 mg.During the experimental period,no abnormalities were observed in all physiological indicators of the mice.No lesions were observed in the heart,liver,spleen,lungs,and kidneys of the KSC 0.5 mg groups compared with negative group.Comparisons of observations and clinical studies shows GIcNAc is safe.Conclusion Kappa-Selenocarrageenan and N-Acetyl-D-Glucosamine can be used as an adjuvant to enhance humoral immune response induced by HAV in mice.
