Objectiv:To observe the effect of Qing Li Yang Yin Huo Xue Decoction on the expression of monocyte chemoattractant protein 1 and macrophage migration inhibitory factor in rats with Ig A nephropathy,in order to explore its regulatory mechanism.Methods:SD rats were divided into normal group,model group,tripterygium wilfordii group,valsartan group,traditional Chinese medicine low and high dose group.The Ig A nephropathy rats were established by using bovine serum albumin,lipopolysaccharide,carbon tetrachloride and castor oil.After successful modeling,every group rats were afforded with corresponding medicine from the eleventh week.At the seventeenth week,urine total protein,serum creatinine and blood urea nitrogen levels of all rats were analyzed.Ig A immune fluorescence was tested by immunofluorescence technology.Pathological changes such as glomerular,renal tubules,interstitial and renal blood vessels in renal tissue were observed by using optical microscope.The m RNA expression of renal MCP-1and MIF were examined by reverse transcription PCR.The correlative analysis was performed with 24-hour urine protein,renal function,renal pathology score,and Ig A immunofluorescence intensity.Results:Compared with the normal group,the UTP,Scr and BUN were significantly increased in the model group and the treatment groups,the expression of MCP-1 m RNA and MIF m RNA in the kidney raised obviously,kidney pathological damage was aggravated,Ig A immunofluorescence was enhanced in the renal tissue,and the difference was statistically significant(P<0.05).Compared with the model group,the UTP,Scr,and BUN of the rats with Ig A nephropathy in each treatment group were significantly lower,the expression of MCP-1 m RNA and MIF m RNA in the kidney significantly decreased,the renal pathological damage was improved,the immunofluorescence of Ig A in the renal tissue of the valsartan group and the high dose traditional Chinese medicine group was reduced,and the difference was statistically significant(P<0.05).Compared with the low dose group and the valsartan group,the high dose group of Qing Li Yang Yin Huo Xue Decoction was more effective in the above mentioned effects(P<0.05).The expression of MCP-1 m RNA and MIF m RNA in kidney of Ig A nephropathy rats was positively correlated with urinary total protein,renal function,kidney pathological changes and Ig A fluorescence intensity,and had the highest correlation with Scr.Conclusion:Qing Li Yang Yin Huo Xue Decoction can reduce the urinary protein excretion of Ig A nephropathy rats,protect renal function,improve the degree of renal pathological injury and reduce the deposition of Ig A immune complexes in the kidney.Its mechanism may be related to the decrease of MCP-1 m RNA and MIF m RNA expression in renal tissue.
