Objective: To discuss the change characteristics of Rho A/ROCK1 signaling pathways in the process of BMSCs differentiated into liver cells.The aim of this study was to identify the new intervention target of BMSCs directional differentiation into liver cells,and then provide better theoretical foundation for clinical transplantation of BMSCs to the treatment of liver cirrhosis.Methods: To culture BMSCs in Vitro,BMSCs were treated with different concentrations of HGF(hepatocyte growth factors)(0ng/ml,10ng/ml,20ng/ml,30ng/ml)in the medium to induce the differentiation of BMSCs.And then we detect the the variation of the cell proliferation with MTT,the contents of AFP and ALB in biological methods,Rho A,ROCK,p-MLC protein expression changes in western blot,in different time(3d,1w,2w,3w).Results:1.The change of cell proliferation showed: Different concentrations of HGF(0ng/ml,10ng/ml,20ng/ml,30ng/ml)induce the differentiation of BMSCs into liver cells,the best concentration of the selected HGF is 20ng/ml to induce the optimal cell proliferation of BMSCs(P<0.05).And the cell proliferation increased with the differention time of BMSCs,the highest cell proliferation is the time when induction BMSCs to 3w(P<0.05).2.The concentration Changes of AFP and ALB showed: BMSCs cultured in 20ng/ml group and 30ng/ml group were not expressed in 3d of AFP.The expression of AFP was beginning to appear in 1w,enhanced in 2w,reduced in 3w.The expression of ALB was beginning to appear of 20ng/ml group and 30ng/ml group in 2w,enhanced in 3w.When the 10ng/ml group cultivated 3w,AFP expressed a little,the expression of ALB was not seen.The expressions of AFP and ALB were not seen in the 0ng/ml group during the culture process.3.The change of Rho A、ROCK1、p-MLC proteins showed:As the concentration of HGF increased,Rho A,ROCK1 and p-MLC protein showed a declining trend(P<0.05),showed concentration dependence.And with the extension of incubation time,Rho A,ROCK1 and p-MLC protein presented a declining trend(P<0.05),showed time dependence.Conclusion: The Rho A/ROCK1 signaling pathway was inhibited in the process of BMSCs differentiation into liver cells induced by HGF,and it had a certain time and concentration dependence.
