| Objectives The aim of this research to explore the feasibility for establishing the osteoporotic osteoarthritis(OA)model of facet joint using female Sprague-Dawley(SD)rats induced by bilateral ovariectomy(OVX),and to explore the effects of salmon calcitonin on cartilage and subchondral bone of lumbar facet joint in this model.Methods Thirty 3-month-old SPF grade female SD rats were randomly divided into three groups with 10 rats in each group.Control group(Sham group),bilateral ovariectomized group(OVX+V group)and bilateral ovariectomized + salmon calcitonin group(OVX+sCT group).Rats in the sham group received sham surgery.Rats in the OVX+V group and OVX+sCT group received bilateral OVX,followed by gavage of normal saline or sCT(16 IU/kg/2day)respectively.All animals were sacrificed at the end point of 12 week of administration,and then lumbar facet joints were harvested and put into 4% formaldehyde.The lumbar facet joints(L4-L5)was prepared to evaluate subchondral bone histomorphometry by Micro-CT.All the specimens were completely decalcified,and then embedded,sliced,safranin fast green staining and immunohistochemical staining.The severity of joint lesion was analyzed by histological score.The expression of ADAMTS-4 and aggrecan were detected by IRS score.The experimental data of each group were analyzed by SPSS 20.0 software.Results 1 The result of safranin fast green staining analyzed by histological score in OVX+V group was higher than that in Sham group,and the difference was statistically significant(P < 0.05).The histological score in OVX+sCT group was lower than that in OVX+V group,but the difference was no statistically significant(P >0.05).2 Micro-CT results showed that bone mineral density(BMD),bone volume fraction and trabecular thickness of both OVX+V group and OVX+sCT group were significantly lower than those in Sham group,and there were statistical differences between the three groups(P < 0.05).Compared with OVX+V group,the values of BMD,BV/TV and Tb.Th of OVX+sCT group increased,and the difference was statistically significant(P < 0.05).3 The results of immunohistochemical staining showed IRS score of ADAMTS-4 protein in OVX+V group was higher than that in Sham group,meanwhile the score of aggrecan protein was lower,and those difference were statistically significant(P < 0.05).The IRS score of ADAMTS-4 in OVX+sCT group was significantly lower than that in OVX+V group,and meanwhile the score of AGG was higher,and those difference were statistically significant(P < 0.05).Conclusions 1 3-month-old SD female rats exhibited lumbar facet joints degeneration at 12 weeks after OVX operation,characterized by irregular cartilage surface,small fissure,and osteoporosis of subchondral bone.2 With administration of sCT,the severity of lumbar articular cartilage lesion was alleviated,the thickness of cartilage was increased,the expression of ADAMTS-4 was decreased,and the expression of AGG was increased,and subchondral bone degeneration was improved,which indicated sCT may be a potential choice for treating osteoporotic OA. |
