| Meat-like Ganoderma lucidum(MGL)is called taisui in Chinese folk and is also recorded in Li Shizhen’s "Compendium of Materia Medica".It’s believed that MGL can be used as food and medicine.Because of the function of prolonging life,MGL is regarded as "the superior product" in "Compendium of Materia Medica".Modern studies shows that tai sui is a Myxomycete complex,a protoplast organism between a creature and a fungus,which possesses original and fungal characteristics.It has multiple pharmacological functions,such as antioxidation,anti-inflammation,improving immunity and so on,but there are few reports on anti-tumor research.To study the function of MGL on anti-tumor,the composition,like polysaccharide and pyrroloquinoline quinone(PQQ),of MGL extract were measured.To study on the antitumor activity of MGL in vitro,MTT method,Hocheset staining method,Annexin V FITC/PI double fluorescence staining,PI single dye flow cytometry were used to detect the effect of MGL extract on HepG2 in cell proliferation,apoptosis morphology,early apoptosis rate and cell cycle.In the meantime,the change of some phenotypic traits,weight and immune organ weight,macrophage phagocytosis and T cell and B cell stimulation index,NK cell activity,ICR tumor-bearing mice spleen immune cell subsets and serum IL-2,IgG and IgM were tested.Analyzing the content of the extract of MGL extract shows that polysaccharide was 165.800±15.011 mg/g,PQQ was 4.198±0.641 mg/g.The content of Ganoderma lucidum is similar to that reported in the literature.cell viability results show that MGL extract has inhibitory effect on HepG2 and LO2 cells,and HepG2 cells survival rate is low compared with LO2 cell In vitro.So,we can conclude that MGL extract can selectively inhibit cell growth.Hoechst staining shows that MGL extract could induce apoptosis.Annexin V FITC/PI double fluorescence staining shows that MGL extract could cause early apoptosis of HepG2 cells.The results of PI single dyeing show that MGL extract could induce cell apoptosis and decrease the amount of DNA.vivo experiments show that MGL extract can slow the tumor-burdened rat weight loss,obviously inhibit the growth of a tumor-burdened mouse tumor,significantly protect the thymus,improve the T cells stimulate index,NK cell activity,CD3 + T lymphocyte subgroup percentage,the content of IL-2.This study provides an experimental basis for the research and development of antitumor drugs of MGL.The above experimental results provide an experimental basis for the artificial cultivation of MGL for anti-tumor drug development. |
