Effect Of Modified Jieyu Recipe On Neuroendocrine And 5-HT1AR Mediated CAMP Pathway In MD Model Rats

Objectives In this study,we observed the behavioral indexes of the rats with perimenopausal depression,Determination of neuroendocrine hormones,such as Estradiol(E2),Follicle stimulating hormone(FSH),Luteinizing hormone(LH).the contents of 5-hydroxy tryptamine 1A receptor(5-HT1AR)and the Protein expression levels on Cyclic Adenosine monophosphate(c AMP),Proteinkinase A(PKA),c AMP-response element binding Protein(CREB)of the depression in Perimenopause model rats,To explore the effect and mechanism of modified Jieyu Recipe on Peri-menopausal depression and to Provide basis for clinical application.Methods In this experiment,72 SD female rats were randomly divided into shamoperated group,model group,modified Jieyu Recipe(low,middle,high group)and western medicine group(Deanxit).The model of perimenopausal depression was established by means of Ovariectomized unite chronic unpredictable mild stimulation and the changes in model behavioral indicators(weight test,sugar-water Preference test,open-field test,forced swim test)were observed before and after treatment.After the completion of the model,drug intervention was Performed,the Chinese medicine group and the western medicine group were administered with modified Jieyu Recipe and Deanxit respectively.The sham-operated group and the model group were given equal volume of distilled water.After the treatment,the levels of serum hormones(FSH,LH,E2)were detected by ELISA.The contents of 5-HT1 AR in hippocampus of rats were detected by immunohistochemical method,the Protein expressions of c AMP,PKA,and CREB in hippocampus were measured by WB method,Statistical analysis of the experimental data was carried out.Expressed as mean ±standard deviation((?) ±s),Single factor analysis of variance(ANOVA)was used to compare the mean of multiple samples,There was statistical significance in using P < 0.05 as the difference.Results 1)On the 21 st day,compared with the sham-operated group,the rats in the model group,modified Jieyu Recipe group and western medicine Deanxit group significantly decreased their weight growth rate,the scores of horizontal and vertical movement,the total fluid consumption,and the consumption of sugar water.The Percentage of sugar water Preference decreased significantly,and the immobility time of forced swimming increased significantly(P<0.05).On the 21 st day of treatment,compared with the model group,the rats in the modified Jieyu Recipe high dose group and the western medicine Deanxit group increased their weight gain,sugar water consumption and sugar water consumption Percentage,and the scores of horizontal and vertical movement in open box test.The time of forced swimming was decreased(P < 0.05).2)Compared with the sham-operated group,the serum E2 level of female rats decreased significantly after castration,and the level of FSH,LH increased significantly(P < 0.05).21 days after treatment,compared with the model group,the level of E2 in the modified Jieyu Recipe group,the high dose group and the western medicine group was significantly increased(P < 0.01)and the FSH、LH level was significantly decreased(P < 0.01).3)In Hippocam Pus japonicus CA1,CA3 area,the number of cells in the model group was significantly decreased,the cell arrangement was loosely arranged,the staining of the cells was light and the staining was uneven.Compared with the sham operation group,The Positive expression of 5-HT1 AR decreased significantly in the model group,the expression level of c AMP,PKA,CREB decreased significantly(P < 0.05).Compared with the model group,The Positive expression of 5-HT1 AR in Tiao Gen Jieyu recipe,high dose group and western medicine group was obviously increased,the expression of c AMP,PKA,CREB Protein was significantly increased(P < 0.05).Conclusions 1)Modified Jieyu Recipe can improve the Performance of perimenopausal depression model rats,suggesting that it has the effect of improving Perimenopausal depression symptoms.2)Modified Jieyu Recipe can improve the levels of endocrine hormones in rats with perimenopausal depression,suggesting that it can Play an antidepressant effect by regulating the hormone level.3)Modified Jieyu Recipecan enhance the c AMP signaling Pathway through 5-HT1 AR to exert antidepressant efficacy and improve depression symptoms in perimeno pause.It is suggested that the 5-HT1AR-mediated c AMP signaling Pathway is one of the targets for regulating the effects of modified Jieyu Recipe.
