| Objective: To pick out the differentially expressed m RNAs and micro RNAs from the m RNA and micro RNA expression profiling of occupational medicamentosa-like dermatitis induced by trichloroethylene(OMLDT)achieved by gene chip technology;the biological roles of differentially expressed genes in OMLDT was explored and the mutual regulating network of micro RNA/m RNA was initially assembled and analyzed,which could provide bioinformation for the further study on the molecular function of micro RNA/m RNA in the OMLDT pathogenesis.Methods: By collecting peripheral venous blood from 30 OMLDT patients and 58 healthy exposers and screening the micro RNA/m RNA expression profile of 8 pair samples by gene chip technology,differentially expressed genes of the mico RNAs and m RNAs were achieved with bioinformatics analysis.Furthermore,the biological function of these differentially expressed genes was dissected through GO and Pathway,of which the differentially expressed micro RNAs were confirmed by q RT-PCR.The mutual regulating network of the micro RNAs and m RNAs was initially assembled and analyzed in order to explore the potential molecular mechanism micro RNA and m RNA involved in the pathogenesis of the OMLDT.Results:(1)Through the screening of the gene chip technology and the analysis of bioinformatics,4898 differentially expressed genes were achieved,of which 2124 genes were up-regulated while 2774 genes were down-regulated;The up-regulated genes were mainly involved in the cell cycle metabolic pathways.On the other hand,the down-regulated genes were mainly joined in the antigen processing and presentation of the T cell receptor signal pathway.These results at least elucidated some molecular events of defense mechanism against TCE.(2)By the similar way,20 micro RNAs with significantly different expression was found,of which 11 micro RNA were up-regulated while 9 micro RNA were down-regulated;Through RT-PCR technology,the relative expression of 4 differentially expressed mico RNAs mi R-320 c,mi R-425-5p,mi R-324-3p and mi R-425-3p were confirmed in the 30 OMLDT patients and 58 healthy exposers,which was consistent with the results of the gene chip screening.The target genes of the 20 micro RNAs were predicted and analyzed by database and bioinformatics.As the result,711 target genes were achieved,of which there were 390 up-regulated genes and 321 down-regulated genes.The formers were mainly involved in cellular signal transduction.On the other hand,the latters mainly joined in the pentose phosphate and aluronic acid pathway.(3)The initial assembly and analysis of the differentially expressed micro RNAs and m RNAs were completed on the STRING website and Ctyoscape,from which 15 key genes were achieved that involved in the immune activating cell and T cell receptor signal pathway.Conclusion: The gene chip technology and bioinformatics methods are applied to screen different micro RNAs and genes in the pathogenesis of OMLDT in this research for the first time,including a regulatory network of different micro RNAs and genes in the events.From micro RNA and m RNA angle,we investigate that the molecular mechanism of medicamentosa-like dermatitis induced by TCE and provide research foundation for the following test. |
