Objective: In this study obesity-related glomerulopathy mice were used to be the object model,by detecting the expression of TLR4 in obesity-related glomerulopathy in renal tissues of mice,to investigate the TLR4 role and significance in the development of obesity-related glomerulopathy.Methods: 16 healthy mice were randomly selected and divided into 2 groups,8 in each group,as the ORG model group and the control group.The model group was given the high-fat diet,and the control group was given the normol diet.The 2 groups were killed on the 12 weeken,taking the renal tissue,paraffin embedded,sliced,HE stained,and Streptomyces anti biotin protein one peroxidase connection(S-P)was used to carry out immunohistochemical staining experiments to detect the expression of TLR4 protein.This study adopts the Image Pro Plus 6.0 Image analysis system for semi-quantitative analysis;The findings were analyzed by using SPSS 17.0 software.Results: After 12 weeks,the body mass of the mice in the ORG model group [(34.08 ± 1.06)g] was significantly increased than the control group(29.39 ± 1.25)g],and the difference was statistically significant(t =-8.09,P<0.05);the results of HE staining under light microscope showed that the glomeruli were obviously hypertrophy in the ORG model group compared with the control group,suggesting that the the ORG model group was successful established;the results of immunohistochemistry showed that in the control group,the expression of TLR4 was very little or not,and the expression of TLR4 in ORG mice increased significantly.Statistical analysis was performed using SPSS 17 software,the difference was statistically significant(U=1.33,P<0.05).Conclusions: Compared with the control group,expression level of TLR4 in kidney of ORG model mice significantly increased.It indicates that the TLR4 may participate in the occurrence and development of ORG and may provide the basis to find out the mechanisim and the approach to treatment.
