Objective: The purpose of this study is to investigate the effect of MG132 on the proliferation of BIU87 and T24 bladder cancer cells,as well as the optimal drug concentration and the optimal drug effect time of MG132 on bladder cancer cells.Methods: Select BIU87 bladder cancer cells in vitro,making the proteasome inhibitor MG132 in accordance with the drug concentration of 0,1,2,3,4,6,8,10 umol/L,action time at 0,1,2,4,8,12,24 and 48 h,effect on BIU87 bladder cancer cells respectively,using CCK8 kits to detect MG132 influence on BIU87 bladder cancer cell proliferation,select the best drug concentration,best drug action time.According to the best drug concentration,with 12 h,24 h,48 h for drug effect time,use MG132 to effect on T24 bladder cancer cell,using CCK8 kit to detect its effect on T24 bladder cancer cell proliferation.Results: CCK8 results showed that compared with the control group,MG132 had a significant inhibitory effect on the proliferation of T24 bladder cancer cells(P < 0.05).When MG132 was effected on BIU87 bladder cancer cells,the drug action showed a concentration dependent relationship.When the concentration was no more than 2μmol/L,the concentration curve of the inhibitory effect on cell proliferation was relatively steep.The inhibitory effect on cell proliferation tends to be stable when the concentration is 2μmol/L.Moreover,the drug effect was time-dependent,and when the treatment time was less than 24 h,the concentration curve of inhibition of cell proliferation was relatively steep.When the processing time was 24 h,the inhibitory effect on cell proliferation gradually leveled off.Conclusion: Proteasome inhibitor MG132 can significantly inhibit BIU87 and T24 bladder cancer cell proliferation activity,and the role of proteasome inhibitor MG132 on bladder cancer cell best drug concentration result in 2μmol/L,the best drug for 24 h.
