Objective To investigate the effect of dexmedetomidine(Dex)on the apoptosis of hepatocyte line HL7702 under hypoxia/reoxygenation(H/R)and its potential molecular mechanism.Methods The HL7702 model of hypoxia-reoxygenation was established.The proliferation of HL7702 cells was detected by CCK8 and Edu.Cell apoptosis was detected by flow cytometry and hoechst staining.The expressions of Bcl,Bax,Caspase3,TLR-4,NF-κB and 1IKB expression were measured by western blot.Results The proliferation of hepatocyte HL7702 was significantly inhibited at 4 hours of hypoxia and 6 hours of reoxygenation.Dex could effectively promote cell proliferation and inhibit apoptosis.After treatment with Dex,the level of Bcl-2 was increased,Bax and Caspase3 were significantly decreased.TLR-4 and NF-κB expression was significantly inhibited by Dex while IKB expression increased.Conclusion Dex can effectively reduce the apotosis of HL7702 hepatocytes,which may be related to the inhibition of TLR-4/NF-κB pathway.
