| ObjectiveTo determine the serum MANF levels in PCOS women and healthy controls,investigate the association of circulating MANF with insulin resistance,reproductive endocrine hormones and glycolipid metabolic characteristics,and evaluate the effect of metformin on the serum MANF levels in PCOS.Methods66 newly diagnosed PCOS women and 40 age-matched healthy women were recruited in our study.Glycosylated hemoglobin,lipoproteins,sex hormones and serum MANF levels were determined.We conducted OGTT and EHC to assess the insulin sensitivity and pancreatic β cells function.Pearson correlation and multiple linear stepwise regression were performed to analyze the association of serum MANF with insulin resistance and other parameters.ResultsThe serum MANF levels in patients with PCOS were significantly lower than healthy controls.Pearson analysis suggested that serum MANF was positively correlated with M values,negatively correlated with HOMA-IR,AUCi/AUCg,BMI,WHR,FAT,LH/FSH,FAI,non-HDL-C and FFA.The multiple linear stepwise regression showed that only M values and FAI were independently related to serum MANF levels.Circulating MANF levels dramatically increased in PCOS women after six-month metformin treatment.ConclusionWe firstly demonstrated that serum MANF levels were decreased in PCOS women,and were negatively correlated with insulin resistance.Circulating MANF levels might be associated with insulin resistance in PCOS women. |
