| Background:Lung cancer is the most common malignancy in the world,which includes two subtupes,small cell lung cancer(SCLC)and non-small cell lung cancer(NSCLC).There are few positive symptoms in early stage lung cancer.Despite the progress of medical technology,we have made great progress in the treatment of malignancies,but the total survival rate of patients with advanced lung cancer is still very low.Therefore,new molecular projects and entities must be identified in order to develop effective strategies for cancer prevention.Long non-coding RNA(lncRNAs)is a linear RNA molecule that has a large number of objective existence in a variety of cells,with a length of more than 200 bases,but does not have the function of encoding protein.Many long non-coding RNA(lncRNA)have been reported to be associated with lung cancer,which is important for the research of lung cancer treatment.According to previous studies,lncRNA CRNDE has been characterized to be implicated in several human cancers.While the function of CRNDE in NSCLC still remains to be explored.Studies have confirmed that lncRNA CRNDE plays a regulatory role in many cancers and affects the prognosis of patients.However,so far,the relationship between CRNDE and lung cancer is still unknown.The aim of this study is to investigate the role of CRNDE in lung cancer in vivo and in vitro.Methods:Part 1.We conducted an in silico analysis with data from TCGA to investigate the association between CRNDE and LC.Kaplan-Meier plotter was used to analyze the function of CRNDE on LC patients’ prognosis.Part 2.Tumor tissues were collected from lung cancer patients who was underwent surgery in Affiliated Hospital of Qingdao University.QRT-PCR was performed to study the expression of CRNDE in lung cancer and analysis the correlations between CRNDE and multiple clinical data.Part 3.A549 lung cancer cell was transfected with pcDNA-CRNDE,and MTT assay was performed to investigate the function of CRNDE on cell proliferation.In vivo assay was performed with nude mice,either.Results:Bioinformatics analysis and QRT-PCR indicated that expression of CRNDE was higher in stage ILC patients(P<0.01),and survival analysis showed that high expression of CRNDE in LC patients links to better prognosis(P<0.05).Detection of QRT-PCR showed a negative correlation amongCRNDE expression,Ki67 expression and tumor size(P<0.01),which indicated that CRNDE is associated with tumor growth in LC.There is no significant correlation between CRNDE level and lymph metastasis(P>0.05).CRNDE overexpression significantly inhibited cell proliferation and tumor growth in vitro and in vivo(P<0.05).Conclusion:In this article,our preliminary data showed that CRNDE is overexpressed in early-stage LC and correlated with better prognosis of LC patients possibly by suppressing cell proliferation. |
