Objective:To investigate the effects of perfluorocarbon by intraperitoneal injection on endoplasmic reticulum stress related proteins(GRP78、PERK、CHOP、Caspase-12)and blood fat(TG、LDL、HDL)in atherosclerosis rats and the pathological changes of aorta of rats.To explore the possible protective effects of PFC on endothelial cells in atherosclerotic rats,to provide a new choice for anti-atherosclerotic therapy.Methods:40 male Wistar rats were selected randomly into 4 groups:normal control group,model control group,low dose of PFC group,high dose of PFC group.In addition to the control group,a high-fat diet was adopted,and intragastriced administration with VD3 in the first three days,and then successively administrate for 12 weeks.In the process of modeling,drug intervention was administered twice a week,the normal control group and the model control group were given intraperitioneal injection of saline,while the PFC treatment groups were given intraperitoneal injection of different doses of PFC.After 12 weeks,to compare the pathological changes of aorta and the serum lipid content in each group,and measure the expressions of GRP78、PERK、CHOP、Caspase12 in aorta tissue by Immunohistochemical method.Results: Compared with the control group,the level of TG、LDL were significantly increased in the model group,while HDL was significantly decreased(P<0.01).Compared with the model control group,the level of HDL was significantly increased(P<0.01),while TG、LDL were significantly decreased(P<0.05)in PFC treatment group.The expressions of GRP78、PERK、CHOP、Caspase12 in aortic tissue were significantly increased in model control group compared those in normal group(P<0.05)。Compared with model control group,the expressions of GRP78 were increased(P<0.05)while the expressions of PERK、CHOP and Caspase12 were decreased in PFC treatment group(P<0.05).Compared with low dose group of PFC,the expressions of GRP78 were decreased(P<0.05)in the high dose of PFC group,while the expressions of PERK、CHOP、Casepase12 with no obvious difference was observed(P>0.05).Compared with model group,the treatment groups inhibited the pathological changes of aorta obviously.Conclusion: A certain dose of PFC may improve the lipid metabolism of aorta rats,to reduce the risk factors for atherosclerosis;By interfering with the expression of ERS related proteins,to protect endothelial cells and prevent atherosclerosis.
