| [Objective] Study the biological reaction of carbon fiber reinforced ultrahigh molecular weight polyethylene materials.[Method] 1.Acute cytotoxicity test: after 24 h,48h and 72 h for mice fibroblasts cultured with the experimental group,negative control group and positive control group respectively,the CCK-8 colorimetric assay was performed to calculate the relative proliferation rate of cells.2.Acute hemolysis test: the experimental group,negative control group and positive control group liquids were mixed with the blood of rabbits and performed water bath and centrifugation,and then each OD value after visual inspection of hemolysis reaction was determined,the rate of hemolysis was calculated.3.Local reaction test and chronic hepatorenal toxicity test after implantation: the experimental materials and control materials were implanted into the gluteus maximus of mice of the experimental group and control group respectively,the blood samples were taken after 1 week,3 weeks and 9 weeks of operation to test ALT,AST,BUN and Cr,and then the liver,kidney and materials were taken to implant into local muscles to perform tissue slice for observation and comparison.4.Acute systemic toxicity test: the intraperitoneal injections for the mice of the experimental group and the control group were performed respectively after 0h,4h,24 h,48h and 72 h,the toxic phenomenon in mice of two groups was observed,and the weight of each mouse was measured.The results were evaluated based on the standard on the basis of the observation and weight measurement.[Result] 1.Acute cytotoxicity test: the relative proliferation rates of cells of the materials at 24 h,48h and 72 h after culture were 103.8%,98.02% and 105.5% respectively.2.Acute hemolysis test: the experimental group and the negative control group didn’t have hemolysis,and the positive control group had the complete hemolysis;the hemolysis rate of materials was 0.73%.3.Local reaction test and chronic hepatorenal toxicity test after implantation: There was no statistical significance of difference between ALT,AST,BUN and Cr of the mice in the two groups at each point-in-time;each tissue slice was compared and observed at each point-in-time,and no obvious difference between the two groups was found.4.Acute systemic toxicity test: no toxic phenomenon in the mice of the two groups at each point-in-time after injection was found,and there was no statistical difference of the weight increment between the mice of the two groups at each point-in-time.[Conclusion] Carbon fiber reinforced ultrahigh molecular weight polyethylene materials have good biological reactivity. |
