| PurposesHomeobox A9 protein is a coding sequence specific transcription regulatory factors,involved in many tumor occurrence or development.B-cell lymphoma-2 protein abnormal protein expression can lead to abnormal expression of HOXA9,influencing the full play of the HOXA9 normal function.This experiment by immunohistochemical method to detect HOXA9 and Bcl-2 protein expression in oral squamous cell carcinoma,to explore its clinical significance,providing a new method for discovering new tumor markers.MethodsThis study collected clinical new biopsy specimens in zhongshan city,guangdong province people’s hospital during April 2015-December 2016.Collected 60 cases of OSCC biopsies specimens and 10 cases of normal oral mucosa tissues,by oral pathology experts confirmed by HE staining.Using immunohistochemical staining for HOXA9 and Bcl-2 protein staining.Adopt double score quantitative integral method of the HOXA9 and Bcl-2 protein percentage of positive cells and tinting strength were analyzed.With SPSS13.0 statistics software Analyze the relationship between HOXA9 and Bcl-2 protein in normal group and its significance in OSCC group of the expression,and the correlation between the two groups of indicators.Results(1)HOXA9 in OSCC positive expression rate is 48.3%(29/60),and normal control group is 80%(8/10);Compared the two groups was statistically significant difference(Z=2.55,P= 0.011,P< 0.05).HOXA9 protein expression of the patients with OSCC independent of sex,age,T,M stage(P > 0.05,no statistical significance).But is associated with lymph node metastasis and pathological grading(P < 0.05,statistically significant).(2)The Bcl-2 protein positive rate is 65%(39/60)in OSCC and normal control group is30%(3/10),compared to similar between the two groups have statistical significance.The Bcl-2protein expression has nothing to do with the patient’s gender,age,TNM staging(P > 0.05,no statistical significance).Related to the pathological grading(P < 0.05,statistically significant).(3)HOXA9 and Bcl-2 expression in OSCC degree negative correlation(r = 0.316,P =0.316,P< 0.05).Conclusion(1)HOXA9 protein expression in OSCC group significantly lower than normal group,and are closely associated with lymph node metastasis,pathologic stage,prompt HOXA9 express lower in play a role in the occurrence and development of OSCC.(2)The Bcl-2 protein expression in OSCC group is significantly higher than normal group,and is closely related to the pathological classification,prompt the Bcl-2 positive expression increased in play a role in the occurrence and development of OSCC.(3)The HOXA9 with the Bcl-2 protein expression of negative correlation in OSCC,both may jointly participate in the occurrence and development of oral squamous cell carcinoma. |
