| Objective: To investigate the expression of scaffolding adapter protein Gab2 in cervical squamous cell carcinoma,to explore the relationship between the expression of Gab2 and the clinicopathological features and prognosis of cervical squamous cell carcinoma,to evaluate the potential clinical value of Gab2 in prognostic markers of cervical squamous cell carcinoma.Methods:We collected 72 cases of pathologically confirmed cervical squamous cell carcinoma tissue by Department of Pathology,Affiliated Hospital of Zunyi Medical College from January 2012 to December 2013,and 20 normal cervical tissues were selected as controls group.The expression of Gab2 protein was detected by immunohistochemistry.At the same time,the clinical and pathological data of the patients were collected and follow-up observations were conducted to investigate the relationship between the expression of Gab2 and the clinicopathological features and prognosis of cervical squamous cell carcinoma.In addition,8 cases of cervical squamous cell carcinoma and normal cervical fresh tissues were collected and the expression of Gab2 mRNA was detected by Real-time quantitative PCR.Results:1.We detected the protein expression of Gab2 in cervical squamous cell carcinoma and normal cervical tissue by immunohistochemical method.The results showed that in 72 cases of cervical squamous cell carcinoma,Gab2 protein over-expression in 42 cases,low expression or negative in 30 cases,the over-expression rate of 58.33%.However the Gab2 protein was weakly expressed in 3 of the 20 normal cervical tissues.The rest were negative,and the overexpression rate was 0.00%.There was a statistically significant difference between the two groups(P <0.001).2.We investigated the expression of Gab2 in cervical squamous cell carcinoma and itsclinical pathological features,the results showed that the protein expression of Gab2 in cervical squamous cell carcinoma and lymph node metastasis was positively correlated(P=0.006).In FIGO stage,with the later stages,the expression rate of Gab2 was higher,but the difference was not statistically significant(P>0.05);The expression of Gab2 was not significantly correlated with age,tumor size,histological grade and FIGO stage(P>0.05).3.Follow-up was performed in 72 patients with cervical squamous cell carcinoma.Two patients were lost to follow-up.The follow-up rate was 97.22%.Among them,the 1-year and 3-year overall survival rates(OS)were 90.28% and 55.56%,respectively.Kaplan-Meier survival analysis showed that the survival rate of patients with high expression of Gab2 was significantly lower than that of patients with low expression of Gab2,but the difference was not statistically significant(p=0.16).4.The m RNA expression of Gab2 in cervical squamous cell carcinoma and normal cervical tissue was detected by Real-time quantitative PCR.The results showed that the expression level of Gab2 in 8 cases cervical squamous cell carcinoma was significantly higher than that in 8 cases normal cervix,and the difference was statistically significant(t=12.27,P <0.005).Conclusion:(1)The expression of Gab2 protein and mRNA in cervical squamous cell carcinoma was significantly higher than that in normal cervical tissue.(2)The expression level of Gab2 was correlated with the lymph node metastasis in patients with cervical squamous cell carcinoma.However,the expression of Gab2 was not significantly correlated with age,tumor size,histological grade and FIGO stage. |
