Objective:This study was designed to investigate whether TLR4-Ab has antagonism effect on the LPS-induced junction proteins injury of intestinal mucosal in mice.Methods:24 adult BALB/C mice were divided into control group,the model group and TLR4 treatment group,with 8 mince in each group.The model group were injected with LPS to establish model of endotoxemia(5mg/kg).TLR4 treatment group were given TLR4 antibody(10g/only),the control group were given 0.9% NaCl instead.The mice were killed after 3h and the distal small intestinal tissue was collected,RT-PCR and immunohistochemistry were used to measure the mRNA and protein expression of TNF-?,NF-?Bp65 and.Claudin1,Occludin,ZO-1.Result:1.On the mRNA and protein expression of NF-?Bp65,the model group was significantly higher than the control group(P<0.05),and the TLR4 treatment group was significantly lower than control group(P<0.05);2.On the mRNA and protein expression of TNF-?,the model group was significantly higher than the control group(P<0.05),and the TLR4 treatment group was significantly lower than control group(P<0.05);3.On the mRNA and protein expression of Claudin1,Occludin and ZO-1,the model group was significantly lower than the control group(P<0.05);Compared with the model group,TLR4 treatment group had significantly higher mRNA expression of Claudin1 and ZO-1,and had significantly higher protein expression of Claudin1,Occludin and ZO-1(P<0.05).Conclusion:1.TLR4-Ab can down-regulate the mRNA and protein expression of NF-?Bp65;2.TLR4-Ab can down-regulate the mRNA and protein expression of TNF-a;3.TLR4-Ab can up-regulate the mRNA expression of Claudin1,and ZO-1,and up-regulate the protein expression of Claudin1,Occludin and ZO-1;4.TLR4-Ab has antagonism effect on the LPS-induced junction proteins injury of intestinal mucosal in mice.
