Exploring the mechanism of bone loss and developing the corresponding prevention still have a great significance.In this research,we started from the experiments in vitro to explore the methods of bone loss simulation and study the effect of mechanical force on bone loss.Our methods utilized the interaction between biological solution and biogenic apatite resulting in calcium elimination from devitalized bone to simulate the bone loss.The device added dynamic load to bone sample by transmitting a mechanical force through a steel plunger which is pressed by a swivel bearing driven by a stepping motor.And an ion-selective electrode was used to evaluate elimination of calcium from a bone during the exposure to biological solution in the setup.Microscope,calcium concentration monitoring and SEM-EDS were used to measure the bone loss of bone sample in vitro.The results from these measurements testified that our methods could be used to simulate bone loss.The level of calcium elimination from a bone during the testing showed the visible dissimilarity for the loaded bone and the unloaded one,the calcium concentration in the solution of loaded bone was remarkable less versus unloaded one.The curve of calcium elimination could be divided into four stages,rapid period,slow period,recover period and stable period.From XRD experiments we found that in the loaded bone versus unloaded,the lattice microstrain parameter revealed a notable increase,while the crystallite size showed only the trend to slight increasing.Based on our data,we anticipated that the bone-loading devices would enhance assessment of mechanical stimulation of bone stability in vitro and,as a result,would have the potential to facilitate a strategy of bone loss preventing.It was the first report on using crystallite size and lattice microstrain to describe the effect of mechanical force on bone loss.The results of this article will explore a new way about the research of bone loss simulation and the prophylaxis and treatment of osteoporosis.
