| Objective:Through Yaotuitong Capsule on rats with transplantation of autologous nucleus pulposus model the content of TNF-αin the serum and expression of phosphorylated p38,p-p38,p65 and IKKβin effect research,Yaotuitong Capsule on the treatment of lumbar disc herniation mechanism,for in-depth study the pathogenesis of LDH.Methods:A total of 70 male Sprague-Dawley(SD)rats were randomly divided into groups(n=10):the blank group,the sham operation group,the model group,the FR(p38mapk)inhibitor group,the Yaotuitong Capsule low dose,medium dose and high dose groups,All rats were made up rat model of lumbar nucleus pulposus prolapse unless blank group.To observe the efficacy of pain-alleviating,using their own front and back control method detect the withdrawal threshold of heat pain stimulation in each group,the level of plasma tumor necrosis factor-α(TNF-α)were taken to observe with enzyme linked immunosorbent assay;The pathological changes of L4-6-6 dorsal root ganglion were observed by HE dyeing;The content of p38,p-p38,p65 and IKKβin the dorsal root ganglia from L4-6level were measured through immunoflurescence staining method.Results:Compared the first day after the modeling with the first day before the model,the thermal pain stimulates withdrawal threshold of the rats in the other groups were decreased except for the blank group rats,and differences were statistically significant(P<0.05).Compared the first day after the modeling with the model building after 7 days and14 days,there were a growing tendency of the thermal pain stimulates withdrawal threshold of the rats,the medium and the low dose groups and the FR inhibitor group.In particular,the effect of high,medium dose group and FR inhibitor group were obvious(P<0.05).Blank group:the morphology of DRG neurons is normal,the nucleus is in the center,the nucleoli and the nuclear membrane are clear,and the cytoplasm is filled with fine particles,and the nerve fibers are arranged in order.Model group:DRG neurons were edemaed obviously,the nucleus skewed and the nuclear membrane fuzzied,disordered arrangement of cells and intercellular space increases,the surrounding satellite cells increased,nerve fiber arrangement chaos and not clear.In the sham operation group,the FR inhibitor group and the low dose,medium dose and high dose groups group,DRG structures were between the blank group and the model group,which improved the optimal morphology of the DRG structure with the FR inhibitor group and the high dose group particularly.There was a significant increase in TNF-αin the serum of rats in the model group,and the differences were statistically significant(P<0.05).Compared with the rats in the model group,the levels of TNF-αin the serum of the FR inhibitor group and the low,medium and high dose group were significantly decreased,and the differences were statistically significant(P<0.05).There were statistically significant differences between the FR inhibitor group,the high,medium dose and low dose group(P<0.05).The content of p38 in DRG in the model group was significantly increased,which was significantly different from that in the normal group(P<0.05).Compared with the model group,the difference between the FR inhibitor group and the high and medium dose group were statistically significant(P<0.05).Compared with the low dose group of,the difference between the high dose group and the FR inhibitor group were statistically significant(P<0.05).The content of p-p38 in DRG in the model group was significantly increased,which was significantly different from that in the normal group(P<0.05).Compared with the model group,the difference between the FR inhibitor group and the high,medium and low dose group were statistically significant(P<0.05).Compared with the the low dose group,the difference between the high dose group was statistically significant(P<0.05).The p65content of DRG in the model group was significantly increased,which was significantly different from the normal group(P<0.05).Compared with the model group,the difference between the FR inhibitor group and the high,medium and low dose group were statistically significant(P<0.05).Compared with the low dose group,the difference between the FR inhibitor group and the high and medium dose group were statistically significant(P<0.05).The content of IKKβin DRG in the model group was significantly increased,which was significantly different from the normal group(P<0.05).Compared with the model group,the difference between the FR inhibitor group and the high dose group were statistically significant(P<0.05).Compared with the low dose group,the difference between the FR inhibitor group and the high dose group were statistically significant(P<0.05).Conclusion:(1)After the transplantation of autologous nucleus pulposus,the right posterior limbs of rats displayed conspicrous hyperalgesic response,the posterior limb’s retracting threshold for thermal stimulation degraded significantly,the content of TNF-alpha in serum increased significantly,and the structural of DRG changed significantly.The model of lunbar root pain produced by transplantation of autologous nucleus pulposus in rats was successfully;(2)Yaotuitong capsule has a certain anti-inflammatory and analgesic effects,its anti-inflammatory effect may be one of the mechanisms of treatment LDH;Yaotuitong capsule can improve the structure of DRG in LDH rat model,and promote repair after DRG injury;(4)The result of immunofluorescence showed that:1.The treatment mechanism of Yaotuitong capsule may be achieved by inhibiting the activation of p38MAPK signaling pathway and NF-κB signaling pathway.2.The p38MAPK signaling pathway and NF-κB signaling pathway may have an interactive effect on the pathogenesis of LDH. |
