Objective: To investigate the relationship between dendritic cell(DC)levels with acute graft-versus-host disease(aGVHD)and without aGVHD after allogeneic hematopoietic stem cell transplantation(allo-HSCT).Methods:The count of plasmacytoid dendritic cells(pDC)and myeloid dendritic cells(mDC)were analyzed by Flow Cytometry on day 30 after allo-HSCT from peripheral blood of 42 patients with hematologic diseases.Results: Univariate and multivariate linear regression analysis indicated that,clinical features(including,sex,age,disease type,conditioning regimen and matched HLA of patients)had no effect on the count reconstitution of pDC and mDC on day 30 after transplantation(P > 0.05).aGVHD,which occurred within 30 days after transplantation,was defined as early aGVHD,and aGVHD occurred from + 31 to + 100 days after transplantation was defined as late aGVHD.On day 30 after transplantation,there was no significant difference in the count of pDC and mDC between the patients with early aGVHD and those without aGVHD(P < 0.05).On day 30 after transplantation,the count of pDC and mDC in patients with late aGVHD was significantly lower than that in those without aGVHD(P < 0.05).Compared with patients who had higher count of mDC on day 30 after transplantation,the patients with lower mDC count had higher incidences of late aGVHD(P <0.05).Conclusion:After allo-HSCT,the reconstitution of DC in peripheral blood on day 30 was correlated with the occurrence of aGVHD.The count of p DC and mDCdeclined markly before aGVHD.There was a negative correlation between the count of mDC and the incidence of aGVHD.
