Objective:Test the expression levels of the metastasis associated gene(MTA-1)and metastasis suppressor gene(23-H1 nm23-H1)in normal cervical tissues,cervical intraepithelial neoplasia(CINII-III)tissues and cervical cancer tissues,respectively,and analyze the relevance of them in cervical cancer tissues and the relationship between their expressions and the clinicopathological features of cervical cancer.To discuss the role of metastasis associated gene(MTA-1)and metastasis suppressor gene(23-H1 nm23-H1)in the occurrence,development,infiltration and metastasis of cervical tissues.Methods:Examine the expression of metastasis associated gene(MTA-1)and metastasis suppressor gene(23-H1 nm23-H1)in normal cervix tissues,cervical intraepithelial neoplasia(CINII-III)tissues and cervical cancer tissues by immunohistochemical staining,analyze their expressions and the relationship between their expressions and the clinicopathological features of cervical cancer withχ~2 test,and analyze the relevance of MTA-1and nm23-H1 in cervical cancer with Spearman method.Results:(1)MTA-1 was mainly expressed in cytoplasm or nucleus,and showed brown or brown granules as positive cells.In normal cervical tissue,cervical intraepithelial neoplasia(CINII-III)tissues expression was low,and it was highly expressed in cervical cancer tissues.(2)The positive expression rate of MTA-1 in normal cervix group,cervical intraepithelial neoplasia(CINII-III)tissues and cervical cancer group were 10%,30%and68.3%respectively.Compared among the three groups,the difference was statistically significant(χ~2=30.853,P<0.05).The positive expression rate of MTA-1 in CIN group was higher than that in normal cervix group,but the difference was not statistically significant(χ~2=3.75,P>0.05).The positive expression rate of MTA-1 in cervical cancer group was significantly higher than that in CIN group and normal cervix group,and the difference was statistically significant(χ~2=27.236,11.903,P<0.05).(3)The expression of MTA-1protein in cervical carcinoma was related to clinical stage,depth of myometrial invasion and lymph node metastasis.The difference was statistically significant(χ~2=8.942,5.640,15.674,P<0.05).However there was no significant correlation to age,histological grade and histological type of cervical cancer(P<0.05).(4)nm23-H1 was also mainly expressed in cytoplasm or nucleus,and showed brown or brown granules as positive cells.In normal cervical tissue,cervical intraepithelial neoplasia(CINII-III)tissues expression was high,and it was low expressed in cervical cancer tissues.(5)The positive expression rate of nm23-H1 in normal cervix group,cervical intraepithelial neoplasia(CINII-III)tissues and cervical cancer group were 96.1%、86.7%、51.7%respectively.Compared among the three groups,the difference was statistically significant(χ~2=24.378,P<0.05).The positive expression rate of nm23-H1 in CIN group was lower than that in normal cervix group,but the difference was not statistically significant(χ~2=0.873,P>0.05).The positive expression rate of nm23-H1 in cervical cancer group was significantly lower than that in CIN group and normal cervix group,and the difference was statistically significant(χ~2=18.225,10.55,P<0.05).(6)The expression of nm23-H1 protein in cervical carcinoma was related to clinical stage,depth of myometrial invasion and lymph node metastasis.The difference was statistically significant(χ~2=5.644,7.033,8.450,P<0.05).But there was no significant correlation to age,histological grade and histological type of cervical cancer(χ~2=0.115,4.205,P>0.05).(7)The expression of MTA-1and nm23-H1 in cervical cancer group has a negative correlation(r=-0.592,p<0.05),which indicate that MTA-1,nm23-H1 has a negative synergistic effect in the occurrence and development of cervical cancer.Conclusion:(1)High expression of MTA-1 protein and low expression of nm23-H1protein may be closely related to malignant biological behavior such as invasion and metastasis of cervical cancer.(2)The abnormal expression of MTA-1 and nm23-H1 may play a role in the occurrence,development,invasion and metastasis of cervical cancer,and they have a negative synergistic effect.
