Objective: To investigate the correlation between refractory asthma patients in Xin Jiang province and single nucleotide polymorphism of rs7701443 and rs10052957 sites in the glucocorticoid receptor gene.Method: According to inclusion and exclusion criteria included in 90 cases,divided into three group: the refractory asthma group,the asthma control group and the healthy control group.Each group has 30 cases.Fasting venous blood was collected in the morning.After anticoagulant,centrifugation,PCR amplification of DNA extract,alkaline phosphatase treatment,single base extension,resin purification,Sequenom Mass ARRAY?SNP detection techniques and MALDI-TOF classification techniques were used to compare the SNP classification of the selected genetic sites.Result: rs7701443-AA/AG/GG genotype frequency respectively in three groups: refractory asthma group 10%,50%,40%,control group 13.3%,46.7%,40%,healthy control group 10%,63.3%,26.7%;accord with Hardy-Weinberg genetic balance.The comparison among any two groups have no statistical significance(P>0.05);rs10052957-AA/AG/GG genotype frequency respectively in three groups: refractory asthma group 3.3%,13.3%,83.3%,control group0%,10%,90%,healthy control group 0%,33.3%,66.7%;accord with Hardy-Weinberg genetic balance,The comparison among refractory asthma group and asthma control group has no statistical significance(P > 0.05),The comparison among refractory asthma group and healthy control group has no statistical significance(P>0.05).But the comparison among asthma control group and healthy control group has statistically significance(P =0.028,P<0.05).GG genotype of asthma control group significantly higher than that of healthy control group,GG genotype may be associated with the development of asthma.Conclusion: Gene mutation and gene polymorphism existed in rs7701443 and rs10052957 of NR3C1.This study found no statistical correlation between rs7701443,rs10052957 gene polymorphism and refractory asthma.rs10052957 gene polymorphism may be related to bronchial asthma,and there may be some correlation with the development of asthma.
