Objective:1.After the unilateral ureteral ligation,observe rats’ renal pathology and the changes of renal function indexes in serum,test whether the copy of renal fibrosis model of rats is successful.2.After injecting Kangfuxin Liquid into renal fibrosis model rats,observe the renal pathology,the changes of renal index in serum and the changes of immunohistochemical TGF-β1,Smad3,Smad7 protein expression.Explore Kangfuxin Liquid’s therapeutic effect on the renal fibrosis model of rats and possible mechanism of effect,and provide new ideas for the clinical treatment of renal fibrosis.Method:Select 108 healthy SD rats,half male and half female,200 + 20 g in weight,and divid into six groups randomly: sham operation group,model group,the groups with Kangfuxin Liquid groups in low,middle and high dose and benazepril group,18 rats in each group.Every 18 rats are divided into group 7d,14 d and 21 d.Except for sham operation group,each group uses UUO model,i.e.after anesthesia,open the abdominal cavity,ligate the left ureter.For sham operation group,after opening the abdominal cavity,separate left ureter but not ligate,then close the abdominal cavity.Administrate each group by gavage after the second day of above modeling: give different concentration Kangfuxin Liquid to the groups with Kangfuxin Liquid in low,middle and high dose,same dose benazepril to benazepril group,same dose of saline to sham operation group and model group.Each of group is once per day,and administered by gavage in 7d,14 d,21d.Kill rats and collect abdominal aortic blood and kidney tissue specimens,to make serum,and test Scr,BUN,β2-MG of serum;Taking renal tissue and staining with HE and Masson,observe the pathological changes in the left kidney;Detect the expression of TGF-β1,Smad3,Smad7 in renal tissue of rats with immunohistochemical staining.Results:(1)The changes of Renal pathology: There are no pathological changes in sham operation group.In the model group andKangfuxin Liquid group in low dose,the renal tubular dilatation,renal interstitial fibrosis and glomerular fibrosis are observed.Other treatment groupsare not as serious as model group.With the increase of time,Model group aggravates,meanwhile other each treatment group reduces;(2)Renal function index: The comparison of Scr,BUN,β2-MG about sham operation group and other groups in different periods(P<0.05);Compared with model group,Kangfuxin Liquid groups in middle,high dose,benazepril group are decreased(P<0.05);The Comparison about Kangfuxin Liquid group in low dosewith model group(P>0.05);The Comparison about Kangfuxin Liquid groups in middle,high dose with benazepril group(P>0.05);With the increase of time,there are no obvious differencesin each group.(3)The expression of TGF-β1,Smad3,Smad7 in renal tissue: Compared the sham operation group with other groups,the TGF-β1,smad3 protein expression of the kidney tissues in different time decreases,however,smad7 protein expression increases(P < 0.05);Compared Kangfuxin Liquid groups in middle,high dose,benazepril group with model group,TGF-β1,smad3 protein expression reduces,but smad7 protein expression increases(P < 0.05);Comparison about Kangfuxin Liquid group in low dose and model group(P>0.05);Comparison about Kangfuxin Liquid group in middle,high dose and benazepril group(P>0.05);With the extension of the time,there are no obvious differencesin each group.Conclusion:(1)Kangfuxin Liquid has obvious improving function on the indexes,such as renal function and pathological of model rat.This fact suggests that it can protect renal function and protest the fibrosis of kidney.This function show dose-effect relationship,especially the function in Kangfuxin Liquid groupsin medium and high dose effect are more apparent.Nevertheless,They don’t show the time-effect relationship.(2)The preventing renal tubulointerstitial fibrosis mechanism of Kangfuxin Liquid maybe play a role through inhibiting the expression of rat’s renal tissue TGF-β1and Smad3,promoting the expression of Smad7,affecting the TGF-β/Smads signal pathway to delay the renal fibrosis.
