Flavonoids are widely found in medicinal plants,has been used for the treatment of various diseases,such as hypertension,diabetes,atherosclerosis and so on.The effective extraction of flavonoids solve the cost of expensive,time-consuming,complicated operation,problems of consumption agent is the key extraction research.Therefore,exploring a green,efficient,economical method is very important.This research takes the flavonoid rich Pueraria,astragalus,Huaihua,baicalin as the model medicine study.The effects of extraction time,extraction solvent,solid-liquid ratio and power factor by single factor combined with Box-Behnken response surface methodology(BBD-RSM)were selected to determine the optimum extraction process of IR,and the use of ultra performance liquid chromatography(UPLC)method to establish a rapid method for simultaneous determination of contents of many kinds of flavonoids in herbs.Finally by infrared assisted method,ultrasonic method,reflux method measuring the content of total flavonoids in different medicinal herbs.Combined with the oxygen radical absorbance capacity(ORAC)and 1,1-diphenyl-2-picrylhydrazyl(DPPH)to evaluate the advantages and disadvantages of traditional method and infrared method,through the content of total flavonoids and antioxidant activity of a more comprehensive and reliable screening process.The results are as follows:(1)Establishment of quantitative UPLC methodUPLC was used to study the mobile phase,chromatographic conditions and methodology.According to the properties of the flavonoids in the medicinal materials,the chromatographic conditions,such as the separation,the peak time,the peak and peak area of the formic acid,the gradient of acetonitrile mobile phase,the best column temperature,the detection wavelength and the flow rate were selected,to screen out the gradient of formic acid and acetonitrile mobile phase with good separation and peak time,peak type and peak area,and the best column temperature,detection wavelength,flow rate and so on.Combined with the results of methodology,four quantitative analysis methods were established.The results of the study were all up to the standard requirements,and the performance of the instrument was good.The infrared assisted method was stable and reliable.The results showed that the content of the effective components of the medicinal materials could be determined and further studied.(2)Infrared assisted extraction and traditional extractionSing the infrared assisted method of the drugs the solid-liquid ratio,extraction time,extraction power of single factor screening and through the BBD-response surface method to optimize the factors,finally the optimum extraction process of total flavonoids extraction.Under the optimum conditions,the content of total flavonoids in each herb was: The content of total flavonoids of Astragalus 0.575mg/g,Radix Puerariae was 34.61mg/g,Sophora japonica was 33.199mg/g,Scutellaria was 174.794mg/g.At the same time with the traditional extraction method(ultrasonic extraction,reflux extraction)extraction of total flavonoids in different Chinese medicinal herbs,the highest content of the corresponding 1.142,40.73,30.217182.695mg/g respectively,the results of different extraction methods in addition to the content of Astragalus,Astragalus is less,the traditional extraction method,the content is about 2 times of IR.(3)Antioxidant activityThe drugs were measured ORAC value and DPPH value in the best infrared assisted extraction conditions of Radix Astragali,radix puerariae,Radix Sophorae,the antioxidant ability of ORAC respectively:6576.203±553,5824.016±575,3785.827 ±518,1603.197±78μmoL/g,the highest DPPH scavenging activity respectively:110.83,32.50,53.44,88.49μg/m L.The highest ORAC value corresponding to the traditional extraction method respectively:3428.050±39,3362.656±284,2983.864±232,2300.137±78 μmoL/g.DPPH value:84.13,64.33,70.2,74.46μg/m L.According to the results,the activity of infrared assisted extraction was higher than that of the traditional extraction method,and the antioxidant activity was 2 times higher than that of the traditional one.In this study,a quantitative analysis method was established for the simultaneous determination of flavonoids by UPLC.The results showed that the four kinds of herbs had different activity.The above research results indicate that the efficiency of infrared assisted extraction was higher than traditional extraction.On the study of the content and activity,the whole is superior to the traditional one,the characteristics of high efficiency,economy,environmental protection,green and so on.It can be used to study the extraction process.
