Bletilla striata(Thunb.)Reichb.f.(Orchidaceae),also known as Baiji,not has been widely used for the treatment of hematemesis.Traditional Chinese Medicine(TCM)holds that it is capable of restraining leakage of blood and stopping bleeding,dispersing swelling and promoting tissue regeneration.In this paper,the research group focuses on the traditional efficacy of Bletilla striata,taking powder characteristics as the research object,and study the hemostasis and wound healing.In a previous study,we found that particle size ranges are the main factor affecting hemostatic outcomes.To improve the hemostatic efficiency of BSMPs,explore its hemostatic mechanism and determine the best size range for hemostasis,we characterized BSMPs into different size ranges(350-250,250-180,180-125,125-75 and < 75 ?m),by means of Scanning electron microscope and Fourier transform infrared spectroscopy(FTIR)in conjunction with physical characterization measurements.We conducted in vivo efficacy studies on rats.Through in vitro blood/BSMP coagulation studies,internal characterizations of blood/BSMP aggregation based on texture analyzer stereoscope and physical property analyses were then used to assess the bioactivity and efficacy of BSMPs of different size ranges.Compared to other BSMPs of different size ranges,BSMPs of 350-250 ?m are most efficient in hemostasis.As powder sizes decrease,the degree of aggregation between particles and hemostatic BSMP effects decline.The BSMP in contact with a bleeding surface locally forms a visible particle/blood aggregate as a physical barrier that facilitates hemostasis.Based on the previous research basis,the research group found that the drying method will affect the quality of white and herbs and its biological activity.To investigate the effects of Freeze-dried Bletilla striata ultrafine powder in animal wound healing and its Physicochemical characterization.Ultrafine powders were produced by using ultra mill.Particle size,specific surface and tap density were used to evaluate the effect of micro-characteristic.Nine New Zealand white rabbit were used to establish rabbit ear trauma model by removed all the cortex(Diameter 1cm).The wound of the left ear was treated by Freeze-dried Bletilla striata ultrafine powder for external application.The wound of the right ear has no treatment.Wound healing rates and healing time were calculated 5,10,15,20 and 30 d after treatment.The tissues were harvested at 10,20 and 30 d,for histological,immunohistochemical.The expression of VEGF was determined by real-time PCR.Wound healing rates,CD31 IOD and the expression of VEGF mRNA were significantly higher in ultrafine powder group than in control group.Histological results showed more well-organized bands of collagen,more fibroblasts and less inflammatory cells in ultrafine powder group compared with control group.Bletilla striata powder in contact with blood can quickly absorb blood,while the powder produces sticky,can be wrapped in blood components to form a polymer,filled in the wound to hemostasis;Bletilla strata rich in mucopolysaccharide is the main cause of its viscosity,powder particles Diameter and surface structure changes will affect the powder adsorption of blood and the effect of powder adhesion;The freeze-dried Bletilla striata ultra-fine powder can improve the wound healing,absorption of wound secretions,to provide a suitable environment for the wound,shorten the inflammation Response time,expression of a large number of vascular endothelial growth factor,promote granulation tissue neovascularization,accelerated wound repair.
