| Objective:With modle establishiment of Radiation-induced lung injury with rats,Toobserve the changes of pulmonary fibrosis detected by section staining and TGF-β1and itsdownstream molecusα-SMA in the lung tissue of the rats treated with Sang Xing Lung-Protection Particles,to find its effects on fibrosis factor.Methods:144 male SD rats weighing 200±20g were randomly divided into blank control group,model group,prednisone group,high,medium and low dose groups of Sang Xing Lung-Protection Particles,each group of 24 rats..Except the blank group,rats were treated with linear accelerator 6MV-X-ray leading to fibrosis model,the dose of each of groups of Sang Xing Lung-Protection Particles was according to the human dose converted into rat equivalent dose.High,medium and low dose were:22.50ml/kg·d,11.25ml/kg·d,5.6ml/kg·d.After 24h of modeling,to give the corresponding drug:blank control group and model group,according to the criteria of 10ml/d normal saline;prednisone group,according to the criteria of 15.60mg/kg/d orally prednisone solution.Eight rats in each group were randomly selected at 24th 28th and 32th weeks;and then the rats were killed and the lung tissue was collected.Masson staining was used to evaluate the degree of fibrosis in lung tissue,and the expression of TGF-β1 andα-SMA was detected by immunohistochemistry.Result:1.Pathological section:The alveolar structure of the lung tissue was clear and no fibrosis at all time points in the blank group.The pathological changes of the pulmonary fibrosis in the model group were more obvious,and the fibrosis of the lung tissue was found at the 16th week Diffuse fibrosis formation;prednisone group and mulberry apricot lung granule group compared with the pathological changes in the model group;2.The expression of TGF-β1,α-SMA and HYP in the blank group were lower than those in the control group(P<0.05).The levels of TGF-β1,α-SMA and HYP in the blank group were lower(P<0.01).Compared with the model group,the levels of TGF-β1,α-SMA and HYP in the prednisone group were lower than those in the blank group(P<0.01)(P<0.01).Compared with the prednisone group,the apricot group was significantly lower than that of the model group(P<0.05).Conclusion:1.Sang Xing Lung-Protection Particles can reduce the degree of pulmonary fibrosis in rats with radioactive pulmonary fibrosis;2.Sang Xing Lung-Protection Particlesr can reduce the levels of TGF-β1,α-SMA and HYP in rats with radiation-induced pulmonary fibrosis. |
