| Objective: To establish the model of cardiomyocytes injury induced by high concentration of palmitic acid and investigate the protective effect of tetrandrine and its mechanism.Methods: Cultured cardiomyocytes were randomly divided into 4 groups: normal group(cardiomyocytes were cultured in DMEM containing 5.5 mmol/L glucose),model group(cardiomyocytes were cultured in DMEM containing 0.2 mmol/L PA),drug delivery group(cardiomyocytes were cultured in DMEM containing 0.2 mmol/L PA and 10 μmol/L Tet)and control group(cardiomyocytes were cultured in DMEM containing 10 μmol/L Tet).The effect on the cell viability of H9C2 cells was evaluated using MTT assay;the expression of Bcl-2,Bax,cleaved caspase-3 were measured by western blot method;the activity of Lactic Dehydrogenase(LDH)in H9C2 cells was detected by LDH kit;the expression of PICK1,IL-6,TNF-α mRNA were measured by RT-PCR.Results: After treatment with PA(0.2 mmol/L)for 24 h,compared with normal group,the cell viability of H9C2 cells was significantly decreased(P<0.01);the expression of Bcl-2 protein in H9C2 cells was decreased(P<0.05);the expression of Bax,cleaved caspase-3 proteins in H9C2 cells were increased(P<0.05 or P<0.01);the expression of PICK1,IL-6,TNF-α mRNA were increased(P<0.05).Compared with the model group,the cell viability was increased(P<0.01)in PA combined with Tet treatment,the expression of Bcl-2 protein in H9C2 cells was increased(P<0.05),and the expression levels of Bax,cleaved caspase-3,PICK1,IL-6,TNF-α mRNA were decreased in Tet treatment(P<0.05 or P<0.01).Conclusion: PA(0.2 mmol/L)can induce H9C2 cells injury after incubation for 24 h,tet may relieve cardiomyocyte injury induced by PA,the mechanism may be related to anti-inflammatory,antioxidant and apoptosis inhibition. |
