Objective:To investigate the effect of Beclin1 on the expression of p-PI3 K 、 p-AKT and p-mTOR in SiHa cells of cervical cancer.Methods:Transfection of Beclin1 gene expression plasmid pcDNA3.1(+)-Beclin1 and interference plasmid pSUPER-Beclin1 into SiHa cells by lipofectamine 2000.Set up the experimental group((1):pcDNA3.1(+)-Beclin1;(2):pSUPER-Beclin1)、negative control group((3):pcDNA3.1(+)no-load group;(4):pSUPER no-load group)、blank control group((5)).qRT-PCR was used to detect the expression of Beclin1 in each group,to extract total RNA at 48 h and the total proteins at 72 h after transfection.Then to investigate the expression of p-PI3 K,p-AKT and p-mTOR at mRNA and protein levels was detected by qRT-PCR and western blot.Results:1.The expression plasmid pcdna3.1(pdna3.1)and the interference plasmid psuper-beclin1 were transfected in SiHa cells,then detected by qRT-PCR.The results showed that the mRNA level of Beclin1 in the overexpression group was significantly increased,and the mRNA level of Beclin1 in the silencing group was significantly decreased,indicating that the transfection was successful.2.After overexpression of Beclin1 in SiHa cells of cervical cancer,the expression of p-PI3 K,p-AKT and p-mTOR in mRNA and protein levels were lower than that innegative control group and blank control group(P < 0.05).3.After silencing the expression of Beclin1 in SiHa cells of cervical cancer,the expression of p-PI3 K,p-AKT and p-m TOR in mRNA and protein levels were higher than that in negative control group and blank control group(p < 0.05).Conclusion:1.After overexpression of Beclin1 in SiHa cells of cervical cancer,the expression of related proteins in the PI3K/AKT/mTOR pathway were decreased at the level of mRNA and protein.2.After silencing the expression of Beclin1 in SiHa cells of cervical cancer,the expression of related proteins in the PI3K/AKT/mTOR pathway were increased at the level of mRNA and protein.
