The Effect Of MiR-224 And Folate On The Formation Of Autophagosome In SiHa Cells

Objective:To investigate the effect of miR-224 and folate on the formation of autophag-osome in Si Ha cells.Methods:1 、 Dividing SiHa cells which in logarithmic growth into five groups: miR-224 mimic group,miR-224 mimic negative control group,miR-224 inhibitor group,miR-224 inhibitor negative control group and blank control group.The expression level of miR-224 in Si Ha cells had been changed by the technology of cell transient transfection.48 hours later,observing the formation of autophagosome in each group.2、Dividing SiHa cells which in logarithmic growth into five groups,different doses of folate were used to interfere with SiHa cells(0.1μg/ml、1.0μg/ml、10μg/ml、100μg/ml、1000μg/ml).And the group of 10μg/ml was the matched group;0.1μg/ml、1.0μg/ml were the low does groups;100μg/ml 、1000μg/ml were the high does groups.72 hours later,observing the formation of autophagosome in each group.Results:1、The amount of autophagosome in miR-224 mimic group was significantly lower than blank control group(P<0.05).2、The amount of autophagosome in miR-224 inhibitor group was significantly higher than blank control group(P<0.05).3、The amount of autophagosome in the low dose of folate group(0.1μg/ml)wassignificantly lower than that of the normal dose of folate group(P < 0.05).The low does of folate group(1.0μg/ml)was lower than that of the normal dose of folate group(P >0.05).4、The amount of autophagosome in the high dose of folate groups(100μg/ml、1000μg/ml)were significantly higher than that of the normal dose of folate group(P <0.05).Conclusion:1、The overpressing miR-224 may be inhibit the formation of autophagosome in SiHa cells,and inhibit the expression of miR-224 may be promote the formation of autophagosome in SiHa cells.2 、 Low dose of folate may be inhibit the formation of autophagosome in Si Ha cells,and the high dose of folate may be promote tthe formation of autophagosome in SiHa cells.
