Chinese medicine has a long history,Its theoretical system comes from the Treatment based on syndrome differentiation of traditional Chinese medicine.Metabonomics is a new discipline developed after genomics,which is used to reveal the phenomena and essence of life activities.development of modern Chinese medicine has provided an opportunity for developing metabonomics technology,making metabonomics an unprecedented development in the field of traditional Chinese medicine.Yinchen wuling powder composed of Yin Chen,Polyporus,Alisma,Atractylodes,tuckahoe,cassia twig six herbs,which is from Zhang Zhongjing's Golden Chamber in the late Eastern Han Dynasty.Main treatment:"damp and hot jaundice,wet and hot,detrimental urination,and thirst."Typhoid and insidious summer-heat evil,Detrimental and thirsty urinating.Wine jaundice,difficulty in micturition,yin jaundice,difficulty in micturition.” Yang Shitai in the "grass hook yuan" Volume 9" Dry yellowing.Dampness.Dryness is powder,especially for the treatment mechanism of yinchen wuling powder.The purpose of this study is to explore the metabolic pathway analysis of three kinds of diseases by yinchen wuling powder,Using gas chromatography and mass spectrometry(GC/MS),liquid chromatography and mass spectrometry(HPLC/MS),linear regression and other analytical methods,to find potential differential metabolites,Verifying its structure with standard products,Construction of metabolic network Atlas.From Potential differential metabolites to find out the mechanism of yinchen wuling powder for treatment jaundice type liver injury,hyperlipidemia and atherosclerosis.Specific research results are as follows:One,Study on the mechanism of anti-icteric liver injury effect on yinchen wuling powder based on metabonomicsIn order to explored the regulation of the jaundice in liver injury in the treatment of yinchen wuling powder.A-naphthyl isothiocyanate causing hepatic injury in rats with jaundice,pathological state of patients with simulated jaundice type liver injury.After modeling,pathological section of rat liver,the liver surface was infiltrated by a large number of inflammatory cells in the model group,and Animal behavior shows the whole body with yellow.GC/MS technique was used to determine the changes in the serum substance in the experimental group and the normal group,the date matrix to SIMCA-P12.0 to have an analysis of PCA and SPLS-DA,screened out the metabolites related to liver injury.Screening out 14 different metabolites:valine,glycine,succinic acid,citric acid,palmitic acid,inositol,linoleic acid,stearic acid,serine,threonine,glyceric acid,malic acid,peanut four acid,cholesterol,MetPA through metabolic pathway enrichment,metabolic network construction and the results show that the yinchen wuling powder the treatment mechanism of the three main metabolic pathways:linoleic acid metabolism,Glyoxylate and Dicarboxylate metabolism,glycine,serine,and threonine metabolism.Two,Study on the mechanism of anti-hyperlipidemia effect on yinchen wuling powder based on metabonomicsIn order to explore yinchen wuling powder effects in the process of treatment in metabolic pathway of hyperlipidemia.A model of hyperlipidemia in rats was given in this study to simulate hyperlipidemia model in humanbody.The pathological section showed that the liver surface of the model group was covered with fat bubbles.The total cholesterol and triglyceride contents in serum were detected by biochemical kit.It was found that the two indexes in the model group were significantly higher than those in the normal group(p<0.05).Pharmacological indicators show the success of the model.The collected serum was analyzed by GC/MS,and the urine was analyzed by HPLC/MS.The date matrix to SIMCA-P12.0 and Metaboanalyst to have an analysis of PCA and OPLS-DA,screened out the metabolites related to hyperlipidemia.Screening out 16 different metabolites: alanine,threonine,glycine,serine,pyroglutamic acid,glutamic acid,corysolic acid,taurine,betaine,l-carnitine,citric acid,oxalosuccinic acid,hydroxypyruvate phosphate,glycerol triphosphate,galacturonic acid,phenylacetylglycine.It was found that the five metabolic pathways most closely related to the regulation of yinchen wuling powder were D-glutamic acid and D-glutamate metabolism,serine,threonine and glycine metabolism,glyoxylate and dicarboxylate metabolism,glyoxylic acid and dicarboxylic acid metabolism,alanine,aspartate and glutamate metabolism.Three,study on the mechanism of anti-atherosclerosis effect on yinchen wuling powder based on metabonomicsIn order to explore yinchen-wuling powder effects in the process of treatment in metabolic pathway of atherosclerosis.A model of atherosclerosis in rats was given in this study to simulate atherosclerosis model in humanbody.The pathological section showed that the liver surface of the model group was covered with fat bubbles,the aortic arch is accompanied by atheromatous precipitation.The total cholesterol and triglyceride contents in serum were detected by biochemical kit.It was found that the two indexes in the model group were significantly higher than those in the normal group(p<0.05).Pharmacological indicators show the success of the model.The collected serum was analyzed by GC/MS,and the urine was analyzed by HPLC/MS.The date matrix to SIMCA-P12.0 to have an analysis of PCA,PLS-DA,OPLS-DA,screened out the metabolites related to atherosclerosis.Screening out 15 different metabolites:glycerol,glycine,serine,inositol,citric acid,linoleic acid,serum creatinine,cholesterol,urinary creatinine,tyrosine,hippuric acid,phenylalanine,3-hydroxybutyric acid,phenylacetyl glycine acid,phenylalanine,3-hydroxybutyric acid,phenylacetylglycine,galacturonic acid,after MetPA enrichment,five metabolic pathways were found to regulate atherosclerosis: linoleic acid metabolism,phenylalanine,tyrosine and tryptophan biosynthesis,tryptophan,threonine and glycine metabolism,phenylalanine metabolism,Glycolate metabolism.The study showed that the mechanism of disease regulation by Chinese herbal medicine yinchen wuling powder covers several metabolic pathways,there are common pathways involved in the three diseases: glyoxylic acid and carboxylic acid metabolism pathway,linoleic acid metabolic pathway,serine/threonine/glycine metabolic pathway,yinchen wuling powder treatmentdiseases by balances energy metabolism,inhibits inflammatory factors and scavenges free radicals levels.The metabolic pathway of yinchen wuling powder was studied by metabonomics,and the metabolic pathways were identified and analyzed in order to provide a theoretical basis for the rational use of yinchen wuling powder in clinical practice.
