Pharmacological Evaluation On Nature And Flavor Of The Alkaloid Subdivision Of Stephania Tetrandra S.Moore

Stephania tetrandra is the dry root of Stephania tetrandra S.Moore.Stephania tetrandra,bitter to taste,cold,in the bladder and lung meridian,has the function of moistening dryness,anti-rheumatic and analgesic.Stephania tetrandra often used to treat disease,such as arthralgia which caused by wind and dampness,edema,beriberi,urine embolism and eczema.Stephania tetrandra as a time-honored TCM,there are still some questions for us to realize the properties and tastes and effectiveness of Stephania tetrandra.This study based on the national 973 plan project called "The theoretical research about the nature of TCM based on the efficacy of alleviating water retention "by Professor Kuang Haixue.In order to explain the material basis for the bitter in flavor,we select the estimated method by means of the core of nature and flavor and the effectiveness evaluation system by the pharmacology indicators(relieving fever,analgesic,anti-inflammatory,anti-rheumatism and so on),to research the ascription of nature and flavor from Stephania tetrandra.In clinical manifestations,Stephania tetrandra often used in the treatment of arthralgia which caused by wind and dampness and largely associated with fever,pain,inflammation.This experiment studies the antipyretic effect,the analgesic action and the anti-inflammatory effect for the alkaloid group of Stephania tetrandra.At the same time,it established bovine type II collagen-induced arthritis rat models,observed the effect of alkaloid group on Rheumatoid Arthurs.The alkaloid group of Stephania tetrandra studies pharmacology by means of the core of nature and flavor,to establish contact between the group,nature and flavor and pharmacological actions,to make sure the material basis for the bitter in flavor.1.Research on antipyretic effect.By using dry yeast to fever of model rats to measure the changes of temperature before and after the making model and infusion drug,study the alkaloid of antipyretic effect.Results show that compared with after building,the studies of total alkaloid can significantly reduce the body temperature of rats in 1,3 hour after the treatment,with significant difference(P<0.05).It can figure out the alkaloid of Stephania tetrandra have an antipyretic effect.2.Research on analgesic action.Using hot plate licking foot and acetic acid twisting methods to measure the number of twist body and mice pain threshold,study the analgesic effects of alkaloid.Results show that the alkaloid can reduce the number of mice body torsion and increase the pain threshold of mice.Among them,the alkaloids of low,medium and high dose group the inhibition rate is 21.04%,32.51%and 50.55%for the pain caused by acetic acid;for the pain caused by hot stimulation can significantly increase the pain threshold(P<0.01).Showed by experiment,the alkaloid of Stephania tetrandra have an analgesic effect.3.Research on anti-inflammatory effects.Using the model of mouse ear swelling by xylene,rats foot swelling induced by carrageen glue to measure auricle swelling and toes swelling rates,and study the anti-inflammatory effects of’ alkaloid.Experimental results showed that the studies of alkaloid can inhibit mouse auricle swelling induced by xylene and carrageen glue induced toes swelling of rats(P<0.01).Among them,the alkaloids of low,medium and high dose group of mouse auricle swelling induced by xylene inhibition rate are 31.79%,38.24%and 31.79%;for carageen glue induced toes swelling of rats,the biggest inhibition rate reached 17.58%,31.39%and 31.39%.It showed that the alkaloid of Stephania tetrandra can inhibit the swelling of acute inflammation,have anti-inflammatory effect.Through the establishment the model of lipopolysaccharide stimulate macrophage RAW264.7 cells in mice,and study the quantity of Stephania tetrandra decoction and its split components released inflammatory mediators----NO,TNF-α and IL-6.The experimental results show that in the mass concentration range of 78.13-312.50 mg/L,the studies of decoction and alkaloid can reduce the release of inflammatory factor NO,TNF-α,IL-6(P<0.01),it has anti-inflammatory effects.4.Research on treating rheumatoid arthritis.Study the effect of alkaloids groups of Stephania tetrandra according to nature and flavor on bovine type Ⅱ collagen induced arthritis rat models.It shows that the alkaloid group has significant anti-rheumatic effects which can reduce four indicators:IL-1,IL-1β,IL-6,TNF-α in serum,improves arthritis index and the swelling degree.The pathological section of arthritis can clearly observe the treatment effects.There is an anti-rheumatic effect of Stephania tetrandra.It has an effect on treating rheumatoid arthritis.The ascription research shows that the effect of bitter in flavor reflects the pharmacological action of relieving fever,analgesic,anti-inflammatory,anti-rheumatism.The alkaloid group of Stephania tetrandra has pharmacological actions of relieving fever,analgesic,anti-inflammatory,anti-rheumatism.It preliminarily testifies that the alkaloid group may be the material foundation of Stephania tetrandra for bitter.
