In nature,Salmonella have a wide range of host,a handful of salmonella has selectivity to the host,the vast majority of people and animals.The host can be a sojourner in mammals,reptiles,birds,insects,and the gastrointestinal tract of human,infection rate among a wide variety of domestic and wild animals was 1%~20%.Salmonella,which can cause gastroenteritis,typhoid fever,sepsis and other diseases,is one of four foodborne pathogenic bacteria.Prevention,detection and eradication of salmonella is very important for public health.Epidemic outbreaks of the bacteria most associated with the consumption of food contaminated by the bacteria,so a quick,simple,sensitive and specific detection methods for prevention and control of this disease is very important.Established KMnF3 of nano-particles and nuclear magnetic resonance coupling antibodies(Nuclear Magnetic Resonance,NMR)combined method for rapid detection of salmonella.This paper coupled antibody KMnF3 Nano-particle with a combination of nuclear magnetic resonance method,used for rapid detection of salmonella.KMnF3 preparation and characterization of nano-particles,modifying,between salmonella and Functionalized Nano-KMnF3 coupling monoclonal antibody preparation-antibody-KMnF3,and NMR of molecular probes,using immune nanoparticles as biosensors,to capture and detect specific pathogens in a sample that is the pooint.And the effectiveness of a new method is assessed.Main results is shown below:1.Using 0.3%formaldehyde inactivated salmonella duck thalli as immunogen preparation-antibody 2C11-F3,antibody was obtained by octanoic acid-ammonium sulfate precipitation and purification of ascites.Preparation of KMnF3 by solvent-thermal method and improvement methods,and modifying it with a PEG with a carboxyl group,obtaining 20 nm KMnF3 particles.And characterization,the elements of K,Mn,and F element atomic ratio of 1:1.02:2.95 composition is very similar,and KMnF3 chemical composition very similar to the XPS,checked compliance with further examination requirements.2.Using EDC/NHSS chemically coupled prepared immune KMnF3,for KnF3 separation effect,different buffers,cell age and other NMR spin-lattice relaxation time of impact and the spin-spin relaxation time to be discussed,buffer solution,eluted with distilled water is better,7h bacterial culture can be detected.3.With the increase of salmonella culture time;spin-lattice relaxation time andspin-spin relaxation time increases.Use this method can detect 103-108 CFU/mL of bacterium detection limit of 103-104CFU/mL.
