| [Objective]:Theory under the guidance of doctors toward design formula,prepared relieving physical fatigue function of health food,pharmaceutical formulations and process conditions to determine,establish quality standards for the preparation of functional evaluation.[Method]:1.The total polysaccharides,the extraction of total flavonoids as the index,orthogonal experiment to optimize the extraction process parameters;2 purification by adsorption clarification to clarity,stability and retention of total flavonoids and total polysaccharides,as an indicator for process optimization;3.The use of traditional wet granulation process for preparing a chewable tablet.chewable tablets of color,taste,hardness,organizational status and other sensory indicators optimal formulation standard screening chewable tablets;4.were identified by thin layer chromatography,UV spectrophotometry spectrophotometric determination of total flavonoids,HPLC Determination of icariin,the establishment of’ quality standards;5.administered by oral gavage the mice low,medium and high dose groups of samples,gavage for 30 days,to the swimming time,blood urea content,liver glycogen reserves,the elimination rate of blood lactate index of "yang huo wu wei" chewable tablets relieve physical fatigue functions were evaluated.[Results]:1 prescription optimum conditions for the extraction process:boiling water 2 times,8 times the amount of water 2h,6 times the amount of water 1.5h;2.Refining optimal parameters are:liquid concentrate ratio of 1:1 1,clarifying agent in an amount of 3-6%,liquid temperature of 70 ℃;3.chewable pharmaceutical formulations as:herbs extract:meringue:lactose:mannitol:dextrin = 1:0.01:0.15:0.15:0.5,with 60%ethanol as a wetting agent;4.TLC for the test.in comparison with the reference drug chromatography chromatography or the corresponding position was the same color fluorescent spots or streaks,no corresponding negative control spots;5.UV spectrophotometric determination of the average content of total polysaccharide chewable 149.21 mg/g;6.The high-performance liquid chromatography,in the range 0.5-2.5μg icariin injection volume and peak area was linear relationship,methodological survey precision,stability and reproducibility RSD values were<2%,the average recovery rate was 98.72%,the average content measured three batches of samples of different batches of icariin was 1.998mg/g;7.functional evaluation showed that the health food has prolong swimming time increased glycogen reserves,reducing the role of the area under the blood lactate curve,increase the amount of blood urea nitrogen was significantly lower than the control group,especially in high doses the effect is more obvious.With increasing doses,relieve physical fatigue in order to enhance the effect of the sample.[Conclusion]The optimized extraction and purification process is stable,viable.chewable molding process is simple and reasonable,the quality standards established good reproducibility can be used for the preparation of quality control,health food prepared according to the method described above can alleviate physical fatigue function. |
