Dendrobium officinale is one of Orchid species,which is one of the rare traditional Chinese herbal medicines.It has functions in tonifying stomach and promoting fluid,and nourishing yin and clearing heat.Also,it plays a role in anti-aging and improving immune function.It has been using to clinically cure diseases such as chronic atrophy gastritis,hypertension,diabetes,cancer.Thus the functional mechanism of active components from D.officinale needs to be identified and further studied.Polysaccharide is one of the main active ingredients in D officinale and closely related with its medicine effects.Studies have suggested that the activity of polysaccharide is probably related to its oligosaccharide composition.Therefore,this thesis has conducted the study of D.officinale polysaccharide and its hydrolyzed product.Extracted polysaccharide was treated with glycosidase.Polysaccharide hydrolyzed products were derivatizated with PMP before HPLC analysis.Polysaccharide hydrolyzed products were used to treat C6 and MPC-11 cell lines in order to detect the effect of enzyme-hydrolyzed products on expression and secretion of immune factors,including 78 k D glucose-regulated protein,interleukin 6,and immunoglobulin.Also,the difference of D.officinale polysaccharides between Zhejiang and Yunnan was analyzed.The results obtained are as follows.1.Different originals of D officinale have different contents of polysaccharide.Polysaccharide content in Zhejiang D officinale is 32.1% while Yunnan D officinale is 33.4%.Treated with different glycosidase,generating different enzymolysed products.Polysaccharides from different originals were treated with same glycosidase,also generating different enzymolysed products.Polysaccharides from different originals have same monosaccharide composition,but different content.2.Polysaccharide products enzymolysed by Cellulase(C),Glucuronidase(G),Xylanase(X)stimulated C6 cell,to secret higher levels of interleukin-6 protein in a dose-independent manner,compared to control.Stimulated mouse MPC-11 cells to express significantly higher levels of GRP78 protein in a dose dependent manner,to secret higher levels of immunoglobulin in a dose dependent manner when treated with polysaccharide enzymolysed product with C and G,compared to control.3.Polysaccharide enzymolysed product with C,G,and X enzyme did not show any cytotoxicity and induction of cell proliferation in C6 and HEK293 cell lines,compared to control.
