Objective: By detecting colorectal cancer patients and healthy subjects in serum markers of oxidative DNA damage in 8-hydroxy-deoxyguanosine(8-OHdG)levels,explore(1)Feasibility of serum 8-OHdG level as the clinical diagnosis of colorectal cancer;(2)Effect of surgical intervention in patients with colorectal cancer on serum levels of 8-OHdG;(3)Whether there are diffrernces in serum 8-OHdG levels in colorectal cancer patients with different TNM stages and different clinical pathological stages;(4)Whether there is a correlation between serum 8-OHdG levels and the common tumor makers CEA or AFP in colorectal cancer.Methods: From 2014 to 2015 in the Mianyang Central Hospital for treatment by the biopsy of colorectal cancer diagnosed 50 cases of preoperative blood samples and taking blood samples after a week.To collect blood samples of normal volunteers 50 cases and blood samples 30 cases in patients with non-tumor intestinal surgery.By enzyme-linked immunosorbent assay(ELISA)determination of target serum 8-OHdG levels,while AFP and CEA determination by chemiluminescence.Statistical analysis:(1)serum 8-OHdG level differences between the significance of colorectal cancer patients and normal control group;(2)significance of the difference between the preoperative and postoperative serum 8-OHdG levels in colorectal cancer patients;(3)Serum levels of 8-OHdG difference between postoperative and preoperative patients with non-tumor bowel surgery.(4)significance of the difference between different T stage,N stage,M stage and different clinical pathological stages in colorectal cancer patients;(5)correlation between serum 8-OHdG levels and the common tumor makers CEA or AFP in colorectal cancer patients.Results: 1 colorectal cancer patients serum 8-OHdG levels(21.52 ± 7.96)was significantly higher than the control group(10.96 ± 3.70),the difference was statistically significant(P = 0.000).2.colorectal cancer patients serum 8-OHdG levels staging difference in the different clinicopathological groups was not statistically significance(P = 0.134);colorectal cancer patients serum 8-OHdG levels in different tumor cell invasion depth(T stage)was no statistically significance(P = 0.456);serum levels of 8-OHdG different lymph node metastasis(N stage)between the groups was not statistically significant(P = 0.143);Because the selected object of study without M1,it can not be analyzed in patients with colorectal cancer statistics 8-OHdG level differences between different M staging.3.Non-tumor bowel surgery patients before surgery and after 1 week of serum 8-OHdG levels were not significantly different(P=0.371).4.colorectal patients after colorectal cancer surgery one week serum 8-OHdG level decreased significantly compared with that before surgery,the difference was statistically significant(P = 0.000).5.Colorectal cancer patients serum 8-OHdG levels and AFP is no significant correlation(P = 0.757);patients with colorectal cancer serum 8-OHdG levels and CEA is no significant correlation(P = 0.023).Conclusions: 1 Colorectal cancer patients serum 8-OHdG levels significantly higher than normal control group,suggesting that serum 8-OHdG potential as a secondary diagnosis of colorectal cancer serological markers.2.colorectal cancer patients serum 8-OHdG levels and colorectal cancer clinical staging,TN stage,independent of the degree of cell differentiation,suggesting that serum 8-OHdG levels may not yet be estimated as tumor progression and prognostic indicator.3.Before non-tumor patients undergoing intestinal surgery and a week after the serum 8-OHdG levels were not significantly different,but one week after colorectal cancer surgery patients serum 8-OHdG levels were significantly decreased compared with that before,but not to normal levels,suggesting surgery treatment of colorectal cancer can affect the serum 8-OHdG levels.4.Colorectal cancer patients serum 8-OHdG levels and common gastrointestinal tumor markers AFP was no significant correlation between the presence of a weak correlation with CEA.
