Objective: To investigate the expression of GRP78 and caspase12 in invasive ductal carcinoma and their clinical significances,to provide experimental evidences for better understanding the biological behavior of invasive ductal carcinoma breast. Methods: Immunohistochemistry was employed to detect the expression of GRP78 and caspase12 in breast invasive ductal carcinoma(n=100),and normal breast tissue(n=100).Abnornal expression of GRP78 and caspase12 and their relationship with clinicopathologic parameters in breast invasive ductal carcinoma was investageted. Results: 1.The positive expression of GRP78 in breast invasive ductal carcinoma and normal breast tissue was 76% and 18%,respectively.The positive expression of GRP78 in breast invasive ductal carcinoma was significantly higher than normal breast tissue(χ~2=67.523,P<0.01).2.The positive expression of Caspase12 in breast invasive ductal carcinoma and normal breast tissue was 74% and 12%,respectively.The positive expression of Caspase12 in breast invasive ductal carcinoma was significantly higher than normal breast tissue(χ~2=78.417,P<0.01).3.There was significantly elevated expression of GRP78 in patients with breast cancer in the late pathological grade,HER2 positive and non-triple receptor negative breast cancer(NTNBC)(P<0.05).The expression of GRP78 was not significantly related with the ages of patients, tumor size,WHO grade, lymthaden trasition and molecular clasification of estrogen receptor(ER),progesterone receptor(PR)and Ki67 antigen(P>0.05).4.There was significantly negative relationship between the expression of Caspase12 and the size of tumor(P<0.05).The expression of Caspase12 was not significantly related with the ages of patients, tumor size,WHO grade, lymthaden trasition and molecular clasification of estrogen receptor(ER),progesterone receptor(PR)and Ki67 antigen(P>0.05).5.There was negative relationship of the expression between GRP78 and Caspase12(r=-0.201,P<0.05)in breast invasive ductal carcinoma.Conclusion:1.GRP78 may be associated with the occurrence of breast cancer, proliferation, and invasion.2.Breast cancer Caspase12 mediated endoplasmic reticulum stress exist in the apoptotic pathway, may be involved in breast cancer cell proliferation/apoptosis.3.GRP78 expressed in three negative breast cancer is lower, but to understand the biological characteristics of three negative breast cancer provides a new direction.
