Bjective: Junctions tight(TJs)is an important function of the normal epithelial cell adhesion,they are mechanicallly connected to cell and the formation of epithelial barrier,maintaining the normal transport between cells and epithelial cell polarity.Tight junction(TJs)as the most important functional structure of cell junctions,claudins(CLDNs)is the most important component of tight junctions,and is involved in the pathogenesis of various diseases.In recent years,the role of claudin in cancer has been widely concerned and studied by the scientific community,but the role and mechanism of its role in the esophageal squamous cell carcinoma has been not reported.To research on the effects of claudin-1 on the extracorporeal proliferation、migration and invasion of the esophageal squamous cell carcinoma(ESCC)cell line Kyse150 by transient interference claudin-1 expression in esophageal squamous cancer Kyse150 cells by SiRNA.Methods :1.Tissue microarray was used to detect the expression of Claudin-1 in cancer tissues and cancer adjacent tissues;2,siRNA transient disturbed of claudin-1 expression in esophageal squamous cell carcinoma cell Kyse150;3,MTT assay and the immunofluorescence detected of siRNA claudin-1 gene expression on proliferation of esophageal squamous cell carcinoma cell Kyse150;4.Flow cytometry detected siRNA gene of claudin-1 expression after cycle in esophageal squamous cell carcinoma cell Kyse150;5,Transwell detected siRNA gene of claudin-1 expression in esophageal squamous cell carcinoma Kyse150 invasion and metastasis impact.Results: 1.Tissue microarray results showed that Claudin-1 in esophageal squamous cell carcinoma(30 cases)in 21 cases with positive expression and negative expression in 9 cases,the positive rate was 70%,in the tissue adjacent to carcinoma(60 cases),12 cases of positive expression in 48 cases of negative expression,the positive rate was 20%,compared with the adjacent tissues,the expression of claudin-1 esophageal squamous cell carcinoma tissues increased significantly(X2=8.743,P=0.002 < 0.05);In esophageal squamous cell carcinoma(30 cases): lymph node metastasis(18 cases),including 16 cases of claudin-1 positive expression,2 cases of negative expression,the positive rate was 88.89%;no lymph node metastasis of esophageal squamous cell carcinoma(12 cases).The positive expression of Claudin-1 in 4 cases,8 cases of negative expression,the positive expression in 33.33% esophageal squamous cell carcinoma,lymph node metastasis,the expression of claudin-1 was significantly higher than that in non lymph node metastasis group(X2=7.826,P=0.003 < 0.05);Esophageal squamous Carcinoma(30 cases),grade I lesions(11 cases)in 8 cases expressed positive(positive rate was 72.72%),grade II lesions(12 case)in 8 cases expressed positive(the positive rate of 66.66%)and grade III lesions in 5 cases expressed positive(positive rate is 71.42%),with different degree of differentiation in squamous cell carcinoma of the esophagus claudin-1 expression had no significant difference,X2=0.278,P = 0.563).The expression of claudin-1 was not related to age and sex.2,MTT assay and immunofluorescence assay showed that: compared with the negative interference group,by SiRNA transient interference of claudin-1 expression in esophageal squamous cell carcinoma cell line Kyse150,can inhibit the proliferation of esophageal squamous cell carcinoma cell line Kyse150,the difference was statistically significant(P < 0.05);3,flow cytometry results showed that: compared with the negative the interference group,transient expression of SiRNA interference Claudin-1 in esophageal squamous cell carcinoma cell line Kyse150,esophageal squamous cell carcinoma cell cycle can be blocked in S phase,the difference was statistically significant(P < 0.05);4,Transwell results showed that: compared with the negative interference group,by SiRNA transient interference of claudin-1 expression in esophageal squamous cell carcinoma in Kyse150 can significantly to promote the invasion and metastasis of esophageal squamous cell carcinoma cell line Kyse150,the difference was statistically significant(P < 0.05).Conclusion:1.Claudin-1 in cancer tissue is high expression in tumor adjacent tissue with low expression;2.Claudin-1 could significantly inhibit the proliferation of esophageal squamous cell carcinoma cell Kyse150;3.Claudin-1 to cell cycle arrest of esophageal squamous cell carcinoma cell Kyse150 in S phase;4.Claudin-1 can obviously inhibit the invasion and metastasis of esophageal squamous cell carcinoma cell Kyse150.
