Objective:Discuss Tau on pulmonary vascular endothelial function,pulmonary vascular remodeling effect and on molecules expression of PKCa-TGF-β1 pathway in lung vascular effect of the rats of PASMCs induced by HYP.Methods:Choosen the male Wistar rats to finish the experiments.Rats in hypoxia(HYP)in different days pulmonary causing artery hypertension were divided into four groups:the normoxia group,HYP 7 days,14 days,and 21 days.Observe the effect of in hypoxia the rats’ general condition;through pathological paraffin section of lung tissue observating the changes of pulmonary vessel wall in hypoxia by H&E staining.Western Blot method we research the impact of in hypoxia protein expression of PCNA.Increased hypoxia days affect the general state of rat pulmonary artery and causing excessive cell proliferation induced pulmonary vascular remodeling.Divided into five groups:normoxic group,hypoxia group,hypoxia+Tau low dose group,middle dose group and high dose group.Observing Tau of HPH rats in general,right ventricular systolic pressure(RVSP),RV/(LV+S);observing the lung tissue pathological paraffin section which the effect of Tau on pulmonary vascular wall by H&E staining;The effect of protein increases of PCNA,PKCa,TGF-β1 expression in hypoxia.We are trying to prove that Tau can be effectively reduced right ventricular systolic pressure and improve right ventricular hypertrophy,and shut down the excessive proliferation of pulmonary vascular cells,and prove that PKCα,TGF-β1 join in the process inhibition of pulmonary vascular remodeling inhibition.We study whether Tau inhibit the proliferation of pulmonary vascular cells by inhibiting PKCα-TGF-β1 pathway activity.Divided into four groups:normal control group,hypoxia group,hypoxia + Tau group,hypoxia + Tau + PKC inhibitor group.Figure out that the influence of Tau expression of TGF-β1.Results:1.Hypoxia can inhibit induce PASMCs,reduced RVSP,right ventricular hypertrophy,Tau of the principle of inhibition of pulmonary artery cells.2.PKCα,TGF-β1 at the same time involved in the inhibitory effect of taurine pulmonary vascular remodeling process.3.After joining the PKCa inhibitor,Tau by inhibiting the expression of PKCa,inhibit the proliferation of pulmonary cells,thereby inhibiting pulmonary vascular remodeling.4.Tau inhibits PKCa after reduce the role of TGF-β1 has also been reduced.Conclusion:Tau inhibit the PAH rats’ RVSP,RV/(LV+S),and the proliferation of pulmonary vascular cells by PKCα-TGF-β1 pathway,thereby inhibiting PASMCs pathological changes in pulmonary vascular remodeling.
