Acute myocardial infarction is the myocardial ischemic necrosis on the basis of coronary artery lesions,the occurrence of coronary artery sharp decline or interrupted,so that the corresponding myocardial severe and lasting acute myocardial necrosis caused by acute ischemia.The disease has a high morbidity and mortality.Early diagnosis is the key.Therefore,the detection of myocardial markers is of vital significance to the early diagnosis of AMI.CK-MB can be used for early diagnosis of AMI.Pathology and imaging studies have clearly established the relationship between CK-MB levels in the blood and infarct size.Currently,CK-MB is also used for monitoring reperfusion after thrombolytic therapy and myocardial reinfarction.CK-MB is the one of the important cardiac biomarkers frequently used in the diagnosis of AMI.point-of-care testing(POCT)devices that use diagnostic immunoassays are rapid and can be used wherever the need arises.ObjectiveA rapid and simple FL-ICA with Eu(Ⅲ)chelate polystyrene microparticles was developed to determine CK-MB,with fast,simple and high sensitivity,high specificity,good reliability and repetition rate characteristics.MethodsDouble antibody sandwich reaction mode was used to the test strip.Based on the permeability and the siphon effect,an aliquot(50 uL)of CK-MB standard or a clinical serum sample was placed on the sample pad of a test strip,and the microspheres labeled CK-MB antibody antigen binding complexes were captured test line in the NC membrane,microspheres labeled rabbit IgG were captured in the control line.In 12min,We used aQcare TRF to detecting the relative peak height(absolute peak height-baseline).We recorded the absolute peak height-baseline of test line relative for HT and the control line for Hc.The HT/HC ratio could be used to quantify CK-MB in the sample.ResultsA linear range was 0.85-100.29 ng/mL for CK-MB,and the LOD was 0.029ng/mL.The intra-and inter-assay coefficients of variation(CV)were both<10%and the average recoveries were from 90.17%-112.63%for CK-MB.The system performed well in interference experiments.Furthermore,a highly significant correlation(r=0.9794,P<0.001)between this methodand the commercially available bioMerieux mini VIDAS system were attained for measuring 120 CK-MB samples.Conclusionthe Eu(Ⅲ)chelate microparticles-based FL-ICA is simple,fast,highly sensitive,reliable,and reproducible for point-of-care testing of CK-MB concentrations in serum.
